Microsatellite markers for Monitipora digitata designed using restriction-site associated DNA sequencing

Montipora digitata is a species belonging to the Acroporidae. In Indo-Pacific region, M. widely distributed and dominant of scleractinian coral in South China Sea, however, there are currently no molecular markers suitable for assessing genetic diversity. Here, restriction-site associated DNA sequencing (RAD-seq) was used isolate characterize polymorphic microsatellite loci. A total 317,361 RAD-tags were obtained using RAD-seq, including 6,778 Primer pairs 106 loci ordered twenty-one loci, that amplified reliably identified. The number alleles per locus 2-7, observed heterozygosity 0.111-0.556 with an average value 0.285, expected 0.105- 0.802, 0.536.
