MicroRNA-Regulated Signaling Pathways: Potential Biomarkers for Pancreatic Ductal Adenocarcinoma
نویسندگان
چکیده
Pancreatic ductal adenocarcinoma (PDAC) is the most aggressive and invasive type of pancreatic cancer (PCa) expected to be second common cause cancer-associated deaths. The high mortality rate due asymptomatic progression clinical features until advanced stages disease limited effectiveness current therapeutics. Aberrant expression several microRNAs (miRs/miRNAs) has been related PDAC thus they could potential early diagnostic, prognostic, and/or therapeutic predictors for PDAC. miRs are small (18 24 nucleotides long) non-coding RNAs, which regulate key genes by targeting their 3?-untranslated mRNA region. Increased evidence also suggested that chemoresistance cells associated with metabolic alterations. Metabolic stress dysfunctionality systems compensate altered status foundation cellular damage. Current data have implicated multiple as hallmarks development, such glutamine redox imbalance, oxidative stress, mitochondrial dysfunction. Hence, both aberrant dysregulation in metabolism can unfavorable effects biological processes, apoptosis, cell proliferation, growth, survival, response, angiogenesis, chemoresistance, invasion, migration. Therefore, these dismal statistics, it crucial develop beneficial strategies based on an improved understanding biology mediators. This review focuses miR-mediated pathways resistance mechanisms evaluates impact alterations

Pancreatic ductal adenocarcinoma: cellular origin, signaling pathways and stroma contribution.
Pancreatic cancer has a very poor prognosis, in part due to its diagnosis at late stages of the disease and to limited response to chemotherapy and radiotherapy. The vast majority of pancreatic cancers are classified as pancreatic ductal adenocarcinomas (PDACs). Despite advances in knowledge on the cellular origin of PDAC or the involvement of signal transduction pathways therein, many question...
متن کاملFibrosis-related miRNAs as serum biomarkers for pancreatic ductal adenocarcinoma
We investigated whether serum microRNAs (miRNAs) could be diagnostic or prognostic markers in pancreatic ductal adenocarcinoma (PDAC). We first identified miRNAs showing altered expression in human pancreatic stellate cells (hPSCs) co-cultured with PDAC cells (Panc-1 and BxPC-3) as compared to hPSCs cultured alone. Among the miRNAs with altered expression, let-7d exhibited reduced expression in...
متن کاملProteomic analyses identify prognostic biomarkers for pancreatic ductal adenocarcinoma
Pancreatic ductal adenocarcinoma (PDAC) is a highly aggressive malignancy. Here we show that shotgun and targeted protein sequencing can be used to identify potential prognostic biomarkers in formalin-fixed paraffin-embedded specimens from 9 patients with PDAC with "short" survival (<12 months) and 10 patients with "long" survival (>45 months) undergoing surgical resection. A total of 24 and 14...
متن کاملPotential prognostic biomarkers in pancreatic juice of resectable pancreatic ductal adenocarcinoma
Despite potentially curative surgery pancreatic cancer has a dismal prognosis. Serum cancer antigen 19-9 (CA 19-9) correlates with tumor burden, resectability and survival in patients with pancreatic ductal adenocarcinoma. Identification of novel biomarkers may facilitate early diagnosis of pancreatic cancer and improve survival. Pancreatic juice is a rich source of cancer-specific proteins ren...
متن کاملCandidate microRNA biomarkers of pancreatic ductal adenocarcinoma: meta-analysis, experimental validation and clinical significance
BACKGROUND The diagnostic and prognostic value of microRNA (miRNA) expression aberrations in pancreatic ductal adenocarcinoma (PDAC) has been studied extensively in recent years. However, differences in measurement platforms and lab protocols as well as small sample sizes can render gene expression levels incomparable. METHODS A comprehensive meta-review of published studies in PDAC that comp...
