MicroRNA-595 promotes osteogenic differentiation of bone marrow mesenchymal stem cells by targeting HMGA2

Purpose: To investigate the effect of miR-595 on osteogenic differentiation bone marrow mesenchymal stem cells (BMSCs).Methods: Human BMSCs were osteogenically differentiated, and protein expression alkaline phosphatase (ALP), osteocalcin (OCN), Runt-related transcription factor 2 (RUNX2) evaluated by western blot. Expression was measured quantitative reverse (qRT-PCR). The viability determined MTT assay. Osteogenic assessed ALP Alizarin red S (ARS) staining. target gene predicted TargetScan analysis validated luciferase activity assay.Results: MiR-595 higher in differentiated than undifferentiated (p < 0.01). ALP, OCN, RUNX2 also upregulated increased BMSCs, mineralized matrix formation, activity. High mobility group AT-hook (HMGA2) lower found to be a miR-595. Overexpression HMGA2 attenuated miR-595-induced increase cell viability, activity, BMSCs.Conclusion: miR-595/HMGA2 axis is involved suggesting that it promising therapeutic for osteoporosis.
