Microarray detection method for pathogen genes by on-chip signal amplification using terminal deoxynucleotidyl transferase

Abstract A microarray detection method based on on-chip signal amplification using terminal deoxynucleotidyl transferase (TdT) was developed to visualize pathogenic genes. Cyclic olefin copolymer (COC) substrate for microarrays treated with oxygen plasma induce hydrophilic surface properties. The capture probe DNA immobilized the COC by UV irradiation. 3ʹ end of modified a phosphate group provide resistance against TdT reaction. Therefore, reaction triggered only when acquired target gene, and biotin-11-deoxyuridine triphosphate (b-dUTP) continuously added gene. Thereafter, streptavidin-conjugated gold nanoparticles (s-AuNPs) tagged poly uridine tails biotin–streptavidin interaction. visual amplified silver enhancement in presence s-AuNPs. usefulness this confirmed analyzing four pathogens allowing their identification.
