Mesenchymal stem cells induce tumor stroma formation and epithelial‑mesenchymal transition through SPARC expression in colorectal cancer

چکیده
Tumor‑stroma interactions serve a crucial role in the development of colorectal cancer (CRC), which secreted protein acidic and rich cysteine (SPARC) has been implicated. Due to between stromal cells [mesenchymal stem (MSCs)], SPARC gene expression is markedly upregulated CRC cells. The present study investigated its potential as biomarker. Specifically, examined association clinicopathological characteristics 42 cases CRC. was associated with T grade, N grade (TNM classification), stage poor prognosis. Furthermore, area fibroblast‑activating protein‑positive staining around increased SPARC‑positive compared SPARC‑negative cases. Proliferation wound healing assays SPARC‑silenced KM12SM [short hairpin RNA (shSPARC)], reduced demonstrated by reverse transcription‑quantitative PCR, revealed that proliferative migratory capacity shSPARC did not differ from wild‑type (WT) However, it when co‑cultured MSCs. in vivo, immunohistological analysis sequencing were conducted an orthotopic implanted mouse model. Tumor growth lymph node metastasis suppressed shSPARC‑transplanted tumors WT‑transplanted tumors, more marked suppression observed following co‑transplantation Immunohistological examination further reaction epithelial‑mesenchymal transition (EMT) co‑transplanted MSCs, these results consistent using extracted tumors. Overall, suggested dependent on interaction induce EMT promote formation tumor microenvironment, suggesting suitability novel target molecule for treatment.

Mesenchymal Stem Cells Trigger Epithelial to Mesenchymal Transition in the HT-29 Colorectal Cancer Cell Line
Background and Objective: Mesenchymal stem cells (MSCs) promote metastasis in colorectal cancer; however, the mechanism underlying this process is not fully understood. Epithelial to mesenchymal transition (EMT) is a key step in tumor acquisition of metastatic phenotype. We aimed to investigate the effect of MSCs on the expression of EMT markers, as well as cancer stem cell markers in HT-29 col...
متن کاملEpithelial-mesenchymal transition: a hallmark in metastasis formation linking circulating tumor cells and cancer stem cells.
The occurrence of either regional or distant metastases is an indicator of poor prognosis for cancer patients. The mechanism of their formation has not yet been fully uncovered, which limits the possibility of developing new therapeutic strategies. Nevertheless, the discovery of circulating tumor cells (CTCs), which are responsible for tumor dissemination, and cancer stem cells (CSCs), required...
متن کاملMesenchymal Stem Cells Induce Epithelial to Mesenchymal Transition in Colon Cancer Cells through Direct Cell-to-Cell Contact1
We previously reported that in an orthotopic nude mouse model of human colon cancer, bone marrow-derived mesenchymal stem cells (MSCs) migrated to the tumor stroma and promoted tumor growth and metastasis. Here, we evaluated the proliferation and migration ability of cancer cells cocultured with MSCs to elucidate the mechanism of interaction between cancer cells and MSCs. Proliferation and migr...
متن کاملB7H1 Expression and Epithelial-To-Mesenchymal Transition Phenotypes on Colorectal Cancer Stem-Like Cells
Cancer stem cells (CSCs) can invade and metastasize by epithelial-to-mesenchymal transition (EMT). However, how they escape immune surveillance is unclear. B7H1 is crucial negative co-stimulatory molecule but little information about whether it works in CSCs. Therefore, we determined the expression of B7H1 and EMT-associated markers in colorectal cancer stem-like cells to investigate a possible...

Pancreatic ductal adenocarcinoma (PDAC) is one of the most lethal cancers with a 5-year survival rate of less than 5%. Moreover, PDAC escapes early detection and resists treatment. Multiple combinations of genetic alterations are known to occur in PDAC including mutational activation of KRAS, inactivation of p16/CDKN2A and SMAD4 (DPC4) and dysregulation of PTEN/PI3K/AKT signaling. Through their...
