The stability of the two-bonded collagen triple helix.
In the triple helical structure of collagen proposed by RAMACHANDRAN and co-workers1, 2 (see ref. 2 for earlier references), there are two N H . . . 0 hydrogen bonds for every three residues in each chain. In the modification of the RAMACHANDRANKARTHA structure proposed by RICI~ A N D C R I C K a, the three chains are pushed slightly farther apart from their positions in this two-bonded structu...

UNDERSTANDING AND ENGINEERING THE COLLAGEN TRIPLE HELIX Matthew Donald Shoulders Under the supervision of Professor Ronald T. متن کاملThioamides in the collagen triple helix.
To probe noncovalent interactions within the collagen triple helix, backbone amides were replaced with a thioamide isostere. This subtle substitution is the first in the collagen backbone that does not compromise thermostability. A triple helix with a thioamide as a hydrogen bond donor was found to be more stable than triple helices assembled from isomeric thiopeptides.
متن کاملMacrocyclic scaffold for the collagen triple helix.
[structure: see text] Three strands of natural collagen are linked by covalent bonds prior to their folding into a triple helix. We report on a synthetic collagen in which the strands are pendent on a rigid macrocyclic scaffold of C(3) symmetry. The scaffold confers substantial conformational stability upon the collagen triple helix and makes its folding independent of concentration, both desir...
متن کاملEffect of 4-Fluoroproline in the X-Position on the Stability of the Collagen Triple Helix
Introduction Collagen consists of three polypeptide chains that fold into a triple helix. Each natural chain contains many repeats of the sequence: XaaYaaGly, in which a third of the Xaa and Yaa residues are (2S)-proline (Pro). The pucker of a proline ring can be influenced by electronegative substituents, such as the hydroxyl in the naturally occurring residue, (2S,4R)-4-hydroxyproline (Hyp) [...
