Mechanical ventilation preserves diaphragm mitochondrial function in a rat sepsis model

Abstract Background To describe the effect of mechanical ventilation on diaphragm mitochondrial oxygen consumption, ATP production, reactive species (ROS) generation, and cytochrome c oxidase activity content, their relationship to strength in an experimental model sepsis. Methods A cecal ligation puncture (CLP) protocol was performed 12 rats while controls underwent sham operation. Half each group were paralyzed mechanically ventilated. We blood gas analysis lactic acid assays 6 h after surgery. Afterwards, we measured ROS activity. also malondialdehyde (MDA) content as index lipid peroxidation, mRNA expression proinflammatory interleukin-1β (IL-1β) diaphragms. Results CLP showed severe hypotension, metabolic acidosis, upregulation IL-1β expression. Compared controls, spontaneously breathing lower force increased susceptibility fatigue, along with depressed consumption production These generation MDA content.
