چکیده

Background and aimsHeterogeneous high-density lipoprotein (HDL) particles, which can contain hundreds of proteins, affect human health disease through dynamic molecular interactions with cell surface proteins. How HDL mediates its long-range signaling functions various types is largely unknown. Due to the complexity HDL, we hypothesize that multiple receptors engage particles resulting in condition-dependent receptor-HDL interaction clusters at surface.MethodsHere used mass spectrometry-based light-controlled proximity labeling strategy LUX-MS a discovery-driven manner decode HDL-receptor interactions.ResultsSurfaceome nanoscale organization analysis hepatocytes endothelial cells using revealed previously known HDL-binding protein scavenger receptor B1 (SCRB1) embedded community, term synapse. Modulating synapse, composed 60 by silencing individual members showed synapse be assembled absence SCRB1 are interlinked. The aminopeptidase N (AMPN) (also as CD13) was identified an member directly influences uptake into primary aortic (HAECs).ConclusionsOur data indicate preformed residing complexes modulate function suggest new theragnostic opportunities.

2H2O-based high-density lipoprotein turnover method for the assessment of dynamic high-density lipoprotein function in mice.

OBJECTIVE High-density lipoprotein (HDL) promotes reverse cholesterol transport from peripheral tissues to the liver for clearance. Reduced HDL-cholesterol (HDLc) is associated with atherosclerosis; however, as a predictor of cardiovascular disease, HDLc has limitations because it is not a direct marker of HDL functionality. Our objective was to develop a mass spectrometry-based method for the ...

High -density lipoprotein cholesterol as a predictor for diabetes mellitus

Background: Diabetes is a prevalent chronic disease around the world. To evaluate risk of diabetes comprehensively, we developed a score model for the risk prediction with HDL-C as a protective factor. Methods: We extracted physical examination data of 2728 individuals. The data contain 18 demographic and clinical variables. To identify the statistical significant feature variables, the backwa...
