چکیده

In eukaryotes, the nuclear envelope (NE) encloses chromatin and separates it from rest of cell. The Linker Nucleoskeleton Cytoskeleton (LINC) complex physically bridges across NE, linking cytoplasmic components. plants, these LINC complexes are beginning to be ascribed roles in cellular functions, including organization, regulation nuclei shape movement, cell division. Homologs core components, KASH SUN proteins, have previously been identified maize. Here, we characterized presumed LINC-associated maize nucleoskeletal proteins NCH1 NCH2, homologous members plant NMCP/CRWN family, MKAKU41, AtKAKU4. All three localized periphery when transiently heterologously expressed as fluorescent protein fusions Nicotiana benthamiana . Overexpression MKAKU41 caused dramatic changes organization periphery, invaginations that stained positive for non-nucleoplasmic markers inner outer NE membranes, ER. severity was altered by connections actin cytoskeleton. maize, appeared share genetic functions with other control shape, stomatal development, pollen viability. Overall, our data show NCH1, characteristic properties interactions components suggestive at impact overall architecture.

Maize (Zea mays L.).

Agrobacterium tumefaciens-mediated transformation is an effective method for introducing genes into maize. In this chapter, we describe a detailed protocol for genetic transformation of the maize genotype Hi II. Our starting plant material is immature embryos cocultivated with an Agrobacterium strain carrying a standard binary vector. In addition to step-by-step laboratory transformation proced...

Early maize (Zea mays L.) cultivation in Mexico: dating sedimentary pollen records and its implications.

A sedimentary pollen sequence from the coastal plain of Veracruz, Mexico, demonstrates maize cultivation by 5,000 years ago, refining understanding of the geography of early maize cultivation. Methodological issues related to bioturbation involved in dating that record combine with its similarity to a pollen sequence from the coastal plain of Tabasco, Mexico, to suggest that the inception of ma...

Chilling responses of maize (Zea mays L.) seedlings: root hydraulic conductance, abscisic acid, and stomatal conductance.

Maize seedling water relations and abscisic acid (ABA) levels were measured over 24 h of root chilling (5.5 degrees C). At 2.5 h into chilling, leaf ABA levels increased by 40x and stomatal conductance (g(s)) decreased to 20% compared with prechill levels. Despite a rapid g(s) response to root chilling, leaf water potential (Psi(L)) of chilled seedlings decreased to -2.2 MPa resulting in a comp...

Characterization of maize (Zea mays) pollen profilin function in vitro and in live cells.

Profilin is a small, 12-15 kDa, actin-binding protein that interacts with at least three different ligands. The 1:1 interaction of profilin with globular actin (G-actin) was originally thought to provide a mechanism for sequestering actin monomers in the cytoplasm. It has recently become clear that the role of profilin in the cell is more complex, perhaps due to interactions with polyphosphoino...

COMBINING ABILITY AND HETEROSIS IN MAIZE (Zea mays L.)

A line × tester analysis was conducted in maize involving 12 lines and 3 testers for grain yield and its components to determine the heterosis as well as general combining ability (GCA) and specific combining ability (SCA) effects. Highly significant genotypic differences were observed indicated wide range of variability present among the genotypes. GCA and SCA variance for yield per plant numb...
