lncRNA APOC1P1-3 promoting anoikis-resistance of breast cancer cells

چکیده
Abstract Background Anoikis resistance plays a critical role in the tumor metastasis by allowing survival of cancer cells systemic circulation. We previously showed that long non-coding RNAs APOC1P1-3 (lncRNA APOC1P1-3) inhibit apoptosis breast cells. In this study, we explored its anoikis resistance. Methods induced two cell lines (MCF-7 and MDA-MB-231) under anchorage-independent culture conditions studied lncRNA effects on apoptosis. Using Dual-Luciferase activity assay, determined whether it specifically binds to miRNA-188-3P. further lung injecting MDA-MB-231 MDA-MB-231-APOC1P1-3-knock-down female BALB/c nude mice. Results found suppressed early these (demonstrated gain or loss their function, respectively) promoted via reducing activated- Caspase 3, 8, 9 PARP. Moreover, target miRNA-188-3p acting as “sponge” block inhibition Bcl-2 (an anti-apoptosis protein). Conclusions Our study supports theory can promote development binding Bcl-2.

Downregulation of LncRNA GAS5 causes trastuzumab resistance in breast cancer
Therapeutic resistance to trastuzumab caused by dysregulation of long noncoding RNAs (lncRNAs) is a major obstacle to clinical management of HER2-positive breast cancer. To investigate which lncRNAs contribute to trastuzumab resistance, we screened a microarray of lncRNAs involved in the malignant phenotype of trastuzumab-resistant SKBR-3/Tr cells. Expression of the lncRNA GAS5 was decreased in...
متن کاملProtein Profiles Associated with Anoikis Resistance of Metastatic MDA-MB-231 Breast Cancer Cells.
Resistance to anoikis, a cell-detachment induced apoptosis, is one of the malignant phenotypes which support tumor metastasis. Molecular mechanisms underlying the establishment of this phenotype require further investigation. This study aimed at exploring protein expression profiles associated with anoikis resistance of a metastatic breast cancer cell. Cell survival of suspension cultures of no...
متن کاملBIM-EL localization: The key to understanding anoikis resistance in inflammatory breast cancer cells
Inflammatory breast cancer (IBC) is a highly metastatic and rare type of breast cancer, accounting for 2-6% of newly diagnosed breast cancer cases each year. The highly metastatic nature of IBC cells remains poorly understood. Here we describe our recent data regarding the ability of IBC cells to overcome anoikis.
متن کاملlncRNA NEAT1 is closely related with progression of breast cancer via promoting proliferation and EMT.
OBJECTIVE Breast cancer (BC) is one of the most common cancers in females. Abnormal proliferation and metastasis are key reasons that cause mortality in BC patients. More and more evidence showed that lncRNA played an important role in the BC development, but the mechanism was not well established. PATIENTS AND METHODS The expression of lncRNA NEAT1 was detected in 40 BC patients by qRT-PCR t...
متن کاملRoles of lncRNA in breast cancer.
Recent systematic genomic studies have revealed a broad spectrum of lncRNAs that are involved in a variety of disease (diseases), including tumor progression, by regulating gene expression at epigenetic, transcriptional and post-transcriptional levels. However, their exact roles of physiological function and the mechanism (mechanisms) of action are yet to be clarified. In breast cancer research...
