Library Preparation Based on Transposase Assisted RNA/DNA Hybrid Co-Tagmentation for Next-Generation Sequencing of Human Noroviruses

Human noroviruses (HuNoVs) are one of the leading causes foodborne illnesses globally. The viral genome is most essential information for source tracing and transmission pattern monitoring. However, whole sequencing HuNoVs still challenging due to sequence heterogeneity among different genotypes low titer in samples. To address this need, study, Transposase assisted RNA/DNA hybrid Co-tagmentation (TRACE-seq) method was established next generation library preparation HuNoVs. Our data demonstrated that almost (>7 kb) could be obtained from all 11 clinical samples tested. Twelve including GI.3, GI.4, GI.5, GI.8, GII.2, GII.3, GII.4, GII.6, GII.12, GII.13, GII.14, GII.21 were involved. Compared with traditional metagenomics preparation, optimized TRACE-seq greatly reduced interference host’s bacterial RNAs. In addition, sequences can assembled by using less raw sufficient depth along genome. Therefore, high versatility reliability, promising attainment. It particularly applicable viruses a mixed complicated host background unable cultured vitro, like utilized study.
