چکیده
Trichodysplasia spinulosa (TS) is a rare hair follicle disorder that develops in immunosuppressed patients and associated with productive infection by the TSPyV polyomavirus. Affected follicles contain an attenuated matrix, expanded compartment of aberrantly proliferating inner root sheath (IRS)-like cells containing viral particles, disorganized mass shaft-like cells. The expression early region (ER) genes encoding T antigens (TAgs) drive cellular proliferation genome replication tightly restricted to matrix IRS-like To probe potential impact TAg on other aspects epithelial biology, we generated bitransgenic iK5;TSER mice harboring Cre-inducible ER could be activated K5-expressing their progeny using tamoxifen. As previously described, TAgs skin adult led development numerous dysmorphic follicles. Additionally, interfollicular tail epidermis was hyperplastic contained small collections TAg+ cells, some which were multinucleated, clear cytoplasm. Many these (Ki67+) strikingly deficient epidermal keratins (K5, K14, K1, K10). Remarkably, subset aberrant ectopically expressed IRS marker trichohyalin hair-specific keratin. Our findings suggest addition primary function driving cell cycle progression needed for replication, may also contribute human pathology modulating differentiation status target

Seroprevalence of Trichodysplasia Spinulosa–associated Polyomavirus
We identified a new polyomavirus in skin lesions from a patient with trichodysplasia spinulosa (TS). Apart from TS being an extremely rare disease, little is known of its epidemiology. On the basis of knowledge regarding other polyomaviruses, we anticipated that infections with trichodysplasia spinulosa-associated polyomavirus (TSV) occur frequently and become symptomatic only in immunocompromi...
متن کاملExpression of keratin K14 in the epidermis and hair follicle: insights into complex programs of differentiation
Keratins K14 and K5 have long been considered to be biochemical markers of the stratified squamous epithelia, including epidermis (Moll, R., W. Franke, D. Schiller, B. Geiger, and R. Krepler. 1982. Cell. 31:11-24; Nelson, W., and T.-T. Sun. 1983. J. Cell Biol. 97:244-251). When cells of most stratified squamous epithelia differentiate, they downregulate expression of mRNAs encoding these two ke...
متن کاملSpecific detection of trichodysplasia spinulosa–associated polyomavirus DNA in skin and renal allograft tissues in a patient with trichodysplasia spinulosa.
BACKGROUND Trichodysplasia spinulosa (TS) is a rare, disfiguring skin condition that affects immunosuppressed patients, universally involving the central face. New data point to the recently discovered TS-associated polyomavirus (TSPyV) as the causative agent. OBSERVATIONS We report a case of TS in a 48-year-old African American man after renal transplant; via polymerase chain reaction and se...
متن کاملDynamics of Lgr6+ Progenitor Cells in the Hair Follicle, Sebaceous Gland, and Interfollicular Epidermis
The dynamics and interactions between stem cell pools in the hair follicle (HF), sebaceous gland (SG), and interfollicular epidermis (IFE) of murine skin are still poorly understood. In this study, we used multicolor lineage tracing to mark Lgr6⁺ -expressing basal cells in the HF isthmus, SG, and IFE.We show that these Lgr6⁺ cells constitute long-term self-renewing populations within each compa...
متن کاملMerkel cell polyomavirus and trichodysplasia spinulosa-associated polyomavirus DNAs and antibodies in blood among the elderly
BACKGROUND Merkel cell polyomavirus (MCPyV) and trichodysplasia spinulosa-associated polyomavirus (TSPyV) are recently found pathogens causing two rare skin disorders, Merkel cell carcinoma (MCC) and trichodysplasia spinulosa (TS). MCC is proportionally common in the elderly and most often is associated with immunosuppression. TS is a folliculocentric infection seen in patients in an immunocomp...
