Kinetics of Odorant Recognition with a Graphene-Based Olfactory Receptor Mimicry

چکیده

Malaria vector mosquito species rely on a handful of specific pheromones for mating; one them, sulcatone (6-methyl-5-hepten-2-one), is also found in human exudation. Therefore, complete understanding the insect’s olfaction, and rapid real-time methods odorant detection, are required. Here, we mimic recognition nerve cells an antenna with synthetic graphene-based bio-electro-interfacial receptor. By this means, obtain kinetics genuine reaction compare them to electro-antennogram data that represent more complex scenario living insect. The odorant-binding proteins OBP 9A 9B only associate their ligands weakly, showing KDs between 2.1 mM 3 mM, while binding depend structural feature cystine knot modulated by local milieu within protein-aided enhancement zone.

Exchanging ligand-binding specificity between a pair of mouse olfactory receptor paralogs reveals odorant recognition principles

A multi-gene family of ~1000 G protein-coupled olfactory receptors (ORs) constitutes the molecular basis of mammalian olfaction. Due to the lack of structural data its remarkable capacity to detect and discriminate thousands of odorants remains poorly understood on the structural level of the receptor. Using site-directed mutagenesis we transferred ligand specificity between two functionally re...

Olfactory receptor neurons use gain control and complementary kinetics to encode intermittent odorant stimuli

Insects find food and mates by navigating odorant plumes that can be highly intermittent, with intensities and durations that vary rapidly over orders of magnitude. Much is known about olfactory responses to pulses and steps, but it remains unclear how olfactory receptor neurons (ORNs) detect the intensity and timing of natural stimuli, where the absence of scale in the signal makes detection a...

Odorant receptor specificities and receptor combinatorials: implications for olfactory coding.

The perception and discrimination of thousands of different odorants by the vertebrate olfactory system results from the activation of specific olfactory neurons within the olfactory epithelium of the nose (reviewed in Buck, 2000; Firestein, 2001). Activity from these cells is then interpreted by the brain to discern the molecular identity of a given odorant stimulus. How is this process of mol...

Odorant response properties of convergent olfactory receptor neurons.

Information about odorant stimuli is thought to be represented in spatial and temporal patterns of activity across neurons in the olfactory epithelium and the olfactory bulb (OB). Previous studies suggest that olfactory receptor neurons (ORNs) distributed in the nasal cavity project to localized regions in the glomerular layer of the OB. However, the functional significance of this convergence ...

Specificity of odorant-evoked inhibition in lobster olfactory receptor neurons.

Lobster olfactory receptor neurons, like those of many animals, use two modes of olfactory signaling, excitation and inhibition to code olfactory information. Inhibition appears to act through two distinct ionic mechanisms. Here we show that neither ionic mechanism is odor-specific, providing further support for the emerging understanding that there are no inhibitory odorants per se, but rather...
