Isothermal Titration Calorimetry as a Method for Analyzing Protein Adsorption in Ion Exchange Chromatography

Ion exchange chromatography is a widely used method for purification of all types biomolecules in current biotechnological downstream processes. Knowledge on the binding behavior proteins provides valuable insight understanding molecular mechanisms protein interactions biological context. However, thermodynamic parameters such as enthalpy and entropy changes that characterize adsorption are still unknown. Isothermal titration calorimetry applications biosciences has gained its merit to study soluble molecules, inhibition, conformational changes, reaction kinetics, adsorption. case adsorption, lot complications arise since usual models solution no longer valid. This work explains detailed methodology obtention enthalpy, Gibbs energy by using ITC together with equilibrium isotherms.
