Isolation and Optimization of Hydrocarbon – Degrading Bacteria
نویسندگان
چکیده
Aims: To isolate and characterize hydrocarbon – degraders in a contaminated soil sample
Study Design: This experiment was carried out under aseptic conditions. The result confirmed by visible spectrophotometer
Place Duration of Study: Department Biological Sciences, Microbiology Programme, Clifford University, Ihie Campus, Owerrinta, Abia State, Nigeria, between May 2021 to July 2021
Methodology: Soil sample with used this from an automobile mechanic workshop. A ten-fold serial dilution made for each sample, after which 1 ml 10-5 dilutions plated using pour plate method incubated at 37 0C 48 hrs. isolates were then grown on different hydrocarbons (crude oil, fuel or spent engine oil) the degradation efficiency spectrophotometer.
Result: isolation degrading bacteria topsoil subsoil samples workshop located Isiala Ngwa North Local Government Area Junction, State out. Four bacterial species isolated collected, subjected degradation/utilization test it observed that two identified as Bacillus Staphylococcus sp able utilize medium more efficiently than other isolates.
Conclusion: Hydrocarbon auto North, State. Two four strains (Bacillus sp), have highest potential use (fuel, crude oil condemn sole carbon source. These strains' abilities suggest they could be effective bioremediation polluted locations. combat contamination caused motor mechanics who carelessly dispose old environment, well general contamination.

متن کاملIsolation and Functional Gene Analyses of Aromatic-Hydrocarbon-Degrading Bacteria from a Polychlorinated-Dioxin-Dechlorinating Process
Aerobic aromatic-hydrocarbon-degrading bacteria from a semi-anaerobic microbial microcosm that exhibited apparent complete dechlorination of polychlorinated dibenzo-p-dioxins/dibenzofurans (PCDD/Fs) were isolated through enrichment and plating culture procedures with dibenzofuran as the model substrate. By 16S rRNA gene sequence comparisons, these dibenzofuran-degrading isolates were identified...
متن کاملEnrichment, isolation, and phylogenetic identification of polycyclic aromatic hydrocarbon-degrading bacteria from Elizabeth River sediments.
The diversity of indigenous bacteria in sediments from several sites in the Elizabeth River (Virginia) able to degrade multiple polycyclic aromatic hydrocarbons (PAHs) was investigated by the use of classical selective enrichment and molecular analyses. Enrichment cultures containing naphthalene, phenanthrene, fluoranthene, or pyrene as a sole carbon and energy source were monitored by denaturi...
