Isolation and Characterization of Rhizobium from Green Gram (Vigna radiata)

چکیده
Nitrogen is a crucial component that acts as building blocks for nucleic acids and proteins. It abundantly present in atmosphere of the earth but plants are not able to easily utilize it. Diazotrophic microorganisms such Rhizobium convert N2 make it available pulse crops. In study, total thirty-four isolates were recovered, from twenty-two with gram-negative-rods biochemically characterized. Most negative congo red (CR) absorption, glucose-peptone, lactose, hofer’s alkaline assays. These have PGP (plant growth promoting) properties solubilisation mineral, hormone, hydrogen cyanide (HCN) productions along antagonistic activity. Plant study revealed SBGR25 treatment has highest germination per cent (98.0 %), shoot (10.3 cm) total-height (16.6 cm), dry weight (5.60 g). Hence, suggests strains could be used promising bioinoculants crop.

Symbiotic effectiveness of bacteriocin producing and non-producing strains of Rhizobium in green gram (Vigna radiata).
Rhizobium strains nodulating green gram [Vigna radiata (L.) Wilczek] were found to produce bacteriocin on modified Bergersen's medium and inhibited the growth of homologous Rhizobium strains. Four bacteriocin producing and four bacteriocin non-producing strains were compared for their effect on nodulation, in planta nitrogenase activity and plant dry weight of green gram. The bacteriocin produc...
متن کاملIsolation and Characterization of Tonoplast from Chilling-Sensitive Etiolated Seedlings of Vigna radiata L.
Tonoplasts were isolated in a high purity from etiolated young seedlings of Vigna radiata L. (mung bean) utilizing a sucrose density gradient system. The excised hypocotyls were homogenized in a sorbitol-buffer system and the 3,600 to 156,000g pellets obtained after the differential centrifugations were suspended in a sorbitol medium and loaded on a linear sucrose density gradient. After centri...
متن کاملIsolation and Partial Characterization of a Subtilisin Inhibitor from the Mung Bean (Vigna radiata).
The subtilisin inhibitor (MBSI-A) from the mung bean (Vigna radiata (L.) Wilczek) seed has been purified to homogeneity. MBSI-A consists of a single polypeptide chain of 119 residues, with a high content of glutamic acid/glutamine, aspartic acid/asparagine, valine, threonine, and proline (19, 12, 10, 9, and 8 residue percent, respectively). MBSI-A is a potent inhibitor of subtilisin Carlsberg, ...
متن کاملCommunity Profiling of Culturable Fluorescent Pseudomonads in the Rhizosphere of Green Gram (Vigna radiata L.)
Study on microbial diversity in the unexplored rhizosphere is important to understand their community structure, biology and ecological interaction with the host plant. This research assessed the genetic and functional diversity of fluorescent pseudomonads [FP] in the green gram rhizophere. One hundred and twenty types of morphologically distinct fluorescent pseudomonads were isolated during ve...
متن کاملRegulation of urease in Bradyrhizobium colonizing green gram (Vigna radiata (L.) Wilczek).
In the present study attempts have been made to characterize urease expression in slow growing Bradyrhizobium strains TAL442 and MO5 which are endosymbionts of green gram (Vigna radiata (L.)Wilczek). It was found that urease activity in vegetative cells of both the strains was inducible unlike their fast growing counterparts. Mode of regulation in TAL442 was governed by presence of ammonia. Ure...
