چکیده

Abstract Glucocorticoid receptor (GR) are known to be expressed in the ovary and glucocorticoids shown exert direct effects on granulosa cell functions. In clinical setting, menstrual abnormality, amenorrhea hypermenorrhea can patients with glucocorticoid excess. On other hand, also used for treatment of PCOS hyperandrogenism. However, reproductive system have not been fully elucidated. present study, we investigated influence follicular steroidogenesis using primary culture rat cells, by focusing ovarian bone morphogenetic proteins (BMPs) acting as a luteinizing inhibitor. Granulosa cells isolated from female immature rats were treated follicle-stimulating hormone (FSH) presence dexamethasone (Dex) serum-free conditions. After Dex 48 h, changes estradiol (E2) progesterone (P4) production cAMP synthesis induced FSH treatments measured ELISA. Total RNAs FSH, BMPs extracted mRNA levels steroidogenetic factors enzymes, BMP receptors Id-1 quantified real-time RT-PCR. Phosphorylation Smad1/5/9 was evaluated Western blotting lysates or absence Dex. As result, it revealed that decreased FSH-induced E2 cells. accordance steroid results, suppressed P450arom expression well By contrast, augmented P4 concentration-dependent manner. found enhance basal P4-synthetic enzymes including P450scc 3βHSD. Of note, activated target gene transcription phosphorylation, particular, BMP-15 among various ligands BMP-2, -4, -6, -7, -9 -15. It increased ALK-6, type-I BMP-15, turn upregulated GR Given acts an inhibitor suppressing FSH-receptor actions, suggested is functionally linked enhancement endogenous leading negative feedback toward overproduction Collectively, elicit differential P4, which actions mutually enhanced

Regulatory role of BMP-9 in steroidogenesis by rat ovarian granulosa cells.

BMPs expressed in the ovary differentially regulate steroidogenesis by granulosa cells. BMP-9, a circulating BMP, is associated with cell proliferation, apoptosis and differentiation in various tissues. However, the effects of BMP-9 on ovarian function have yet to be elucidated. Here we investigated BMP-9 actions on steroidogenesis using rat primary granulosa cells. BMP-9 potently suppressed FS...

Effects of fenvalerate on steroidogenesis in cultured rat granulosa cells.

OBJECTIVE This study was designed to examine the in vitro effects of fenvalerate on steroid production and steroidogenic enzymes mRNA expression level in rat granulosa cells. METHODS Using primary cultured rat granulosa cells (rGCs) as model, fenvalerate of various concentrations (0, 1, 5, 25, 125, 625 micromol/L) was added to the medium for 24 h. In some cases, optimal concentrations of 22(R...

Regulation of Apoptosis and Steroidogenesis in Ovarian Preovulatory Granulosa Cells

Only a minute fraction of ovarian follicles present in a foetal ovary will complete the path to ovulation. The majority of the follicles will instead undergo atresia, a hormonally controlled apoptotic process. Apoptosis occurs throughout follicle development, with an extensive reduction in the number of follicles present at birth. Interestingly, this drastic reduction is not present among preov...

Modulation of steroidogenesis in human ovarian granulosa cells during aging.

OBJECTIVE To investigate the in vitro effect of endothelin-1 (ET-1) on the steroid production (progesterone [P] and estradiol [E2]) by cultured human granulosa cells (GCs) during aging. MATERIAL AND METHODS Human ovarian GCs and granulosa-luteal cells (GLCs) were isolated from ovaries of female patients (young and premenopausal) undergoing surgery for non-ovarian benign gynecological conditio...

Signaling Mechanism of Fsh and Tgfβ1 in Stimulating Steroidogenesis in Rat Ovarian Granulosa Cells

Previous studies demonstrate that transforming growth factor β1 (TGFβ1) augmented FSH-induced progesterone production in rat ovarian granulosa cells. The post-receptor signaling relaying FSH action rests mainly on cAMP followed by protein kinase A (PKA)-dependent and/or PKA-independent pathway possibly involving phosphatidylinositol-3-kinase (PI3K). This study was conducted to investigate the r...
