Internal validation for analytical and stochastic thresholds in the GlobalFiler system

Interpretation of crime stain profiles is one the challenging tasks forensic scientists. Before using probabilistic genotyping system, scientists should interpret analytical and stochastic thresholds. According to guidelines presented by Scientific Working Group on DNA Analysis Methods, these thresholds shall be based supported applicable internal validation studies. In this study, we performed study our typing system determine A total 350 samples 11 negative controls were amplified GlobalFilerTM PCR Amplification Kit with 30 cycles products then analyzed a SeqStudio Genetic Analyzer. As result, threshold was set 110 RFU, which has high sensitivity obtain almost full 0.063 ng DNA, specificity not detect most pull-up drop-in peaks. The 890 RFU allowing 1% probability allelic drop-out logistic regression. These are useful for interpreting in GlobalFiler system.
