Interleukin-5 Potentiates Sulfidopeptide Leukotriene Production by Human Eosinophils

Interleukin-5 (IL-5) has been shown to be a selective eosinophil growth and differentiation factor. In the present study, the effect of recombinant human IL-5 on human eosinophil sulfidopeptide leukotriene production was investigated. IL-5 did not affect leukotriene synthesis in unstimulated eosinophils. However, IL-5 potentiated leukotriene synthesis by eosinophils stimulated with serum treate...

Leukotriene generation by eosinophils

Horse eosinophils purified to greater than 98% generated slow reacting substance (SRS) when incubated with the calcium ionophore A23187. On a per cell basis, eosinophils generated four to five times the SRS produced by similarly treated horse neutrophils. Eosinophil SRS production was inhibited by 5,8,11,14-eicosatetraynoic acid and augmented by indomethacin and arachidonic acid, suggesting tha...

Analysis of the survival of mature human eosinophils: interleukin-5 prevents apoptosis in mature human eosinophils.

We and other groups have previously shown that interleukin-5 (IL-5) maintained the viability of mature eosinophils in an in vitro liquid culture system. Mature eosinophils did not proliferate but their survival was maintained in the presence of IL-5. Using this culture system, we investigated the mechanism of IL-5-mediated survival. In the absence of human IL-5 (hIL-5) mature eosinophils succum...

Interleukin-5 inhibits glucocorticoid-mediated apoptosis in human eosinophils.

Glucocorticoids (GCs) represent one of the most effective treatments for eosinophilmediated inflammatory diseases such as asthma. GCs act through the GC receptor, leading to proinflammatory cytokine suppression and a reduction in the number of inflammatory cells including eosinophils and T cells. However, the benefits of GCs have been limited by their side effects and the presence of GC resista...

Pneumolysin potentiates production of prostaglandin E(2) and leukotriene B(4) by human neutrophils.

Exposure to pneumolysin (8.37 and 41.75 ng/ml) caused a calcium-dependent increase in the generation of prostaglandin E(2) and leukotriene B(4) by both resting and chemoattractant-activated human neutrophils in vitro. These interactions of pneumolysin with neutrophils may result in dysregulation of inflammatory responses during pneumococcal infection.
