Inter- and Intra-Continental Genetic Variation in the Generalist Conifer Wood Saprobic Fungus Phlebiopsis gigantea

چکیده

Phlebiopsis gigantea (Fr.) Jülich is a well-known generalist conifer wood saprobe and biocontrol fungus used in several world countries to prevent stump infection by tree pathogenic Heterobasidion fungal species. Previous studies have reported the presence of regional continental genetic differentiation host-specific fungi, but such for saprobes as P. has not been often studied or demonstrated. Additionally, little information exists on distribution this western North America. The main purposes study were: (I) assess California, (II) explore variability at intra inter-continental levels (III) analyze phylogeographic relationships between American European populations. Seven loci (nrITS, ML5–ML6, ATP6, RPB1, RPB2, GPD TEF1-α) from 26 isolates coniferous forests diverse geographic different hosts were analyzed together with 45 GenBank sequences. One hundred seventy-four new sequences generated using either universal specific primers designed study. mitochondrial ML5–ML6 DNA ATP6 regions highly conserved did show differences any isolates. Conversely, ITS, TEF1-α variable among samples. Maximum likelihood analysis strongly supported presences two subgroups within species without congruence partition, suggesting retained ancestral polymorphisms. RPB1 RPB2 separated ones, while locus samples eastern ones. This reports California first time DNA-based confirmation identifies older genetically distinct subspecific groups, well three differentiated lineages species: one Europe, America latter presumably including individuals rest identified here indicates that should be (or vice versa), and, likewise, commercially available forests. lack host specificity was documented field survey further reinforces need only use local fungus, given exotic genotypes broad microbe may easily spread permanently alter microbial biodiversity native forest ecosystems.

Physiological and molecular analysis of the interaction between the conifer pathogen,Heterobasidion annosum s.l. and Phlebiopsis gigantea

Prophylactic stump treatment with Phlebiopsis gigantea has been used for a long time in the biological control of the conifer pathogen, Heterobasidion annosum sensu lato. However, the mechanisms underlying the biocontrol process are poorly understood. A total of 64 isolates of P. gigantea from different geographical regions were screened for fungal traits (growth rate, enzyme production, wood d...

Inter- and intra-specific genetic variation in Fusarium.

Genetic variation occurs at all levels across the genus Fusarium. In some cases such variation has been used to define species, and in others to describe populations or lineages. When amplified fragment length polymorphisms (AFLPs) are evaluated, strains in different species usually share at least 60% of the fragments and those in different species 40% of the fragments, or less, with isolates s...

Analysis of the Phlebiopsis gigantea Genome, Transcriptome and Secretome Provides Insight into Its Pioneer Colonization Strategies of Wood

Collectively classified as white-rot fungi, certain basidiomycetes efficiently degrade the major structural polymers of wood cell walls. A small subset of these Agaricomycetes, exemplified by Phlebiopsis gigantea, is capable of colonizing freshly exposed conifer sapwood despite its high content of extractives, which retards the establishment of other fungal species. The mechanism(s) by which P....

Purification by Immunoaffinity Chromatography, Characterization, and Structural Analysis of a Thermostable Pyranose Oxidase from the White Rot Fungus Phlebiopsis gigantea.

A moderately thermostable pyranose oxidase (PROD) was purified to apparent homogeneity with a yield of 71% from mycelium extracts of the white rot fungus Phlebiopsis gigantea by an efficient three-step procedure that included heat treatment, immunoaffinity chromatography, and gel filtration on Superdex 200. PROD of P. gigantea is a glycoprotein with a pI between pH 5.3 and 5.7. The relative mol...
