چکیده
High temperatures exacerbate the ammonia inhibition of anaerobic digestion coupled with methanogenesis. The methane production by has been observed in other studies. However, underlying mechanism is not well understood and requires further investigation. This study explored effect stress on archaeal 16S rRNA transcripts thermophilic digester sludge. Different ammonium concentrations were checked for their influence methanogenic rate hydrogen accumulation. Quantitative PCR was used to compare changes total expression. A Monte Carlo permutation test within redundancy analysis (RDA) adopted exploring relationship between environmental variables transcripts. results showed that increase concentration, decreased accumulation occurred. genes copy numbers significantly treatments higher (7 10 g NH4+-N/L), but did change much at lower (3 NH4+-N/L) compared 0 NH4+-N/L treatment. RDA revealed most variables, including methane, except formate, correlated community structure activity rather than genes. composition hydrogenotrophic methanogen Methanothermobacter dominated all incubations. It exhibited sensitivity should be responsible under conditions. Our findings suggested transcripts, genes, are key indicators activity.

Inhibitory effects of ammonia on methanogen mcrA transcripts in anaerobic digester sludge.
Methanogens in anaerobic ammonia-rich digesters show differential responses to ammonia stress. The mechanism for this is poorly understood. In the present study, we determined the rates of methane production, the composition of methanogen mcrA (the gene coding for the alpha subunit of methyl-coenzyme M reductase) and their transcripts in response to ammonium addition in the anaerobic sludge ret...
متن کاملAnalysis of ammonia monooxygenase and archaeal 16S rRNA gene fragments in nitrifying acid-sulfate soil microcosms.
The present study describes the occurrence of a unique archaeal ammonia monooxygenase alpha subunit (amoA) gene in nitrifying acid-sulfate soil microcosms at pH 3.5. The soil was collected from an abandoned paddy field in Thailand. Microcosms were incubated in the dark at 30°C for 372 days with the following three treatments: addition of ammonium sulfate solution once a month (I) or once a week...
متن کاملCFD simulation of anaerobic digester with variable sewage sludge rheology.
A computational fluid dynamics (CFD) model that evaluates mechanical mixing in a full-scale anaerobic digester was developed to investigate the influence of sewage sludge rheology on the steady-state digester performance. Mechanical mixing is provided through an impeller located in a draft tube. Use is made of the Multiple Reference Frame model to incorporate the rotating impeller. The non-Newt...
متن کاملEffects of Anaerobic Digester Sludge Age on Odors from Dewatered Biosolids
Dewatered sludge cakes from bench scale anaerobic digesters operated at mesophilic temperatures and solids retention times (SRT) ranging from 10 days to 40 days were evaluated for their sulfur odor generation potential. Extracellular Polymeric Substances (EPS) extraction for digested sludge samples was also carried out. Static headspace sulfur gas analysis was successfully used to compare diffe...
متن کاملThe Transition Stage in the Start-up of an Anaerobic Sludge Digester
The start up
dynamics of a one-million and two-hundred-thousand liter anaerobic digester of municipal waste treatment sludge was studied. In the digester, mixing was accomplished by circulation of the produced biogas at a rate of 0.13 m3 per hour of the digester volume per hour. During the nine-month study, no serious phenomena of foaming, getting sour, temperature fall, etc. were observe...
