Inherently confinable split-drive systems in Drosophila

چکیده
Abstract CRISPR-based gene-drive systems, which copy themselves via gene conversion mediated by the homology-directed repair (HDR) pathway, have potential to revolutionize vector control. However, mutant alleles generated competing non-homologous end-joining (NHEJ) resistant Cas9 cleavage, can interrupt spread of elements. We hypothesized that drives targeting genes essential for viability or reproduction also carrying recoded sequences restore endogenous functionality should benefit from dominantly-acting maternal clearance NHEJ combined with recessive Mendelian culling processes. Here, we test split (sGD) systems in Drosophila melanogaster are inserted into required ( rab5 , rab11 prosalpha2 ) fertility spo11 ). In single generation crosses, sGDs variable efficiencies and display sex-biased transmission. multigenerational cage trials, follow distinct drive trajectories reflecting their differential tendencies induce target chromosome damage and/or lethal/sterile mosaic Cas9-dependent phenotypes, leading inherently confinable outcomes.

Multiple meiotic drive systems in the Drosophila melanogaster male.
The behaviour of two "meiotic drive" systems, Segregation-Distorter (SD) and the sex chromosome sc(4)sc(8) has been examined in the same meiocyte. It has been found that the two systems interact in a specific way. When the distorting effects of SD and sc(4)sc(8) are against each other, there is no detectable interaction. Each system is apparently oblivious to the presence of the other, gametes ...
متن کاملMeiotic Drive in Natural Populations of Drosophila
EIOTIC drive has been defined as a force, potentially capable of altering M gene frequencies in natural populations, which somehow depends upon the nature of the meiotic divisions; specifically, when the meiotic divisions are such that the two kinds of gametes from a heterozygote are produced in a ratio different from 1 : 1 ( SANDLER and NOVITSKI 195 7). There have been reported numerous cases ...
متن کاملNocturnal Male Sex Drive in Drosophila
Many behaviors and physiological processes including locomotor activity, feeding, sleep, mating, and migration are dependent on daily or seasonally reoccurring, external stimuli. In D. melanogaster, one of the best-studied circadian behaviors is locomotion. The fruit fly is considered a diurnal (day active/night inactive) insect, based on locomotor-activity recordings of single, socially naive ...
متن کاملExternal noise control in inherently stochastic biological systems.
Biological systems are often subject to external noise from signal stimuli and environmental perturbations, as well as noises in the intracellular signal transduction pathway. Can different stochastic fluctuations interact to give rise to new emerging behaviors? How can a system reduce noise effects while still being capable of detecting changes in the input signal? Here, we study analytically ...
متن کاملLinearly independent split systems
An important procedure in the mathematics of phylogenetic analysis is to associate, to any collection of weighted splits, the metric given by the corresponding linear combination of split metrics. In this note, we study necessary and sufficient conditions for a collection of splits of a given finite set X to give rise to a linearly independent collection of split metrics. In addition, we study ...
