Influence of plant growth regulators on morphogenic response, biomass and camptothecin production in the callus cultures of Chonemorpha fragrans (moon) Alston

In vitro morphogenic response of mature seed embryo-derived callus cultures Chonemorpha fragrans was studied using solid and liquid Murashige Skoog medium amended with cytokinins or their combinations naphthalene acetic acid at 0.5 mg L-1. The plant growth regulators (PGRs) combination concentrations tested could not stimulate organogenesis after three subcultivations the on same PGRs-amended when cultivated in but, formation observed. Evaluation biomass camptothecin production showed that PGRs influenced CPT yield cultures. alkaloid various explants 3-4 weeks old axenic seedlings higher roots (0.019% CPT) followed by embryos (0.0053%), cotyledons (0.0039%), hypocotyls (0.0024%) leaves (0.0017%), no significant difference observed from induced explants. Camptothecin lower than due to extracellular leaching effect alkaloid. Amount synthesized also varied PGR type concentration cultivation medium, association production. Results present study suggest offer alternative tissue source for enhancement through biotechnological approaches, application large-scale conserve ever-decimated natural population medicinal woody climber CPT.
