Influence of extract derived cell cultures of broccoli against osteoporosis in ovariectomized rats

This study was conducted to enhance broccoli secondary metabolites; using plant biotechnology techniques and tissue culture as well evaluate the therapeutic effect of extract against osteoporosis in ovariectomized (OVX) rats. Cell technique implemented order achieve produce polyphenols from leaf explants broccoli. The high-performance liquid chromatography (HPLC) used polyphenolic profiles ethanolic extracts for cell cultures compared dried inflorescences (cultivated plant). However, total phenol flavonoid contents were determined by spectrophotometric technique. Induction performed via ovarectomy operation where rats divided into 5 groups (10 each). Group 1: Sham operated control group remaining 12 weeks model. 2: Ovariectomized osteoporotic positive group. Groups 3-5: rat daily received 17‐β‐estradiol (10 µg/kg), suspension ethanol (300 & 600 mg/kg) orally. obtained results showed that Murashige Skoog basal nutrient medium (MS) fortified with 2.25 µM 2,4-Dichlorophenoxyacetic acid (2,4-D) 0.55 kinetin (kin.) best calli production. Addition methionine enhanced production which identified HPLC. Administration elevated uterine weight, high density lipoprotein-cholesterol estradiol 2 (E2) serum levels reduced lipid profile, liver kidney functions, osteocalcin procollagen type 1 N-terminal propeptide (TPINP). In conclusion, may be a promising sources polyphenol flavonoids treatment bone loss damage osteoporosis.
