In Vitro Regeneration of Stevia (Stevia rebaudiana Bertoni) and Evaluation of the Impacts of Growth Media Nutrients on the Biosynthesis of Steviol Glycosides (SGs)

A plant tissue culture protocol from stevia was optimized for the production of planting materials and natural sweetener, rebaudioside A. The highest survivability (88.90% ± 5.55) explants achieved at 15 30 days after initiation (DACI) on Murashige Skoog (MS) media by sterilization with 30% Clorox (5 min) 10% (10 min), respectively. Supplementation MS 0.50 mg/L 2,4-Dichlorophenoxyacetic acid (2,4-D) 0.10 zeatin produced 50% callus DACI while 1.50 2,4-D increased to 76.67%. shoot proliferation per 10.00 6-benzyl amino purine (BAP) in (5.80) (12.33). longest shoots 4.31 cm 6.04 DACI, respectively, were using BAP (10.00 mg/L) 1.00 naphthalene acetic (NAA). (0.50 strength) induced 2.86 6.20 roots 3.25 7.82 long Stevia grown 0.25 accumulated concentration (6.53%), which correlated expression level its biosynthetic gene uridine-diphosphate-dependent (UDP)-glycosyltransferase (UGT76G1).
