In Vitro Induction and Identification of Polyploid Amorphophallus muelleri Blume Plants by Colchicine Treatment

This research aimed to induce in vitro and identify polyploidy plantlets of Porang (Amorphophallus muelleri Blume) after colchicine treatment. In shoot base explants porang were cultured on MS medium containing different concentrations 30, 60, 90, 120 mg/l for four weeks while the control was without colchicine. After weeks, all treated untreated multiplied regenerated into plantlets. Flow cytometry chromosome count employed determine plantlet’s ploidy level number, respectively. The result showed that inhibited explant growth, formation, plantlet regeneration. Increasing concentration reduces capacity number shoots Of 19 from colchicine-treated explants, nine mixoploid. Percentage mixoploid with 30 higher than 60 Mixoploid plants are mixed diploid, tetraploid, octoploid cells.
