In vitro cancer models as an approach to identify targetable developmental phenotypes in cancer stem cells

چکیده

Abstract Cancer therapeutics are often highly toxic to the patient, and they elicit rapid resistance in tumour. Recent advances have suggested a potential new way which we may improve on this, through two important concepts: (1) that multitudinous pathway alterations converge limited number of cancer cellular phenotypes, (2) these phenotypes depend reactivation developmental processes only minimally active adult tissues. This provides rationale for pursuing an approach ‘drugging phenotype’ focussed targeting reactivated from embryonic development. In this concepts paper, cover recent developments their implications development can avoid patient toxicity acquired resistance. We then propose vitro tumour models provide experimental identify target specific at play, with focus stem cells drive progression spread. Ultimately, aim is cells, essential progression. Therapeutically could represent treats whilst avoiding or acquisition therapeutic

Cancer stem cells: therapeutic targets

Cancer stem cells (CSCs) have been identified as rare cellular populations in many cancers, including leukemia and solid tumors. This minor subpopulation of cancerous cells is immortal tumor-initiating cells which thought to be responsible for cancer initiation, progression, metastasis, recurrence and drug/radiation resistance. Low proliferative rate, high self-renewing capacity, differentiatio...

Investigating the role of signaling pathways and cancer stem cells in esophageal cancer with a therapeutic approach

Esophageal cancer (EC) is the sixth main cause of cancer death worldwide. Important genes associated with esophageal cancer include FOXO3, AKT, and GSK3β. Excessive FOXO3 expression inhibits the proliferation of cancer cells. The expression of AKT is involved in controlling cell growth in tumors. GSK3β activity is higher in cancer tissues. Given the effective role of cancer stem cells (CSCs) in...

Virotherapy as an approach against cancer stem cells.

It has been hypothesized that cancers originate from a small population of cells with stem cell-like characteristics, including self-renewal and pluripotency. Such tumor-initiating cells, also referred to as cancer stem cells, are thought to account for relapses following seemingly successful treatments, because their slow turnover and capacity for expelling anti-tumor drugs leaves them untouch...

Gold nanoparticles as a radio-sensitizer of colon cancer cells at high megavoltage energies: An In-Vitro study

Introduction: In the point of physical view, there are no significant differences between tumor and normal tissues during radiation therapy. Radio-sensitizers have a key role to address the issue. Exploiting high atomic number, gold nanoparticles (GNPs) have been introduced as novel radio-sensitizers and have shown promising result in the field. Owing to high mass attenuation c...

Reprogramming to developmental plasticity in cancer stem cells.

During development and throughout adult life, sub-populations of cells exist that exhibit phenotypic plasticity - the ability to differentiate into multiple lineages. This behaviour is important in embryogenesis, is exhibited in a more limited context by adult stem cells, and can be re-activated in cancer cells to drive important processes underlying tumour progression. A well-studied mechanism...
