In Vitro Alpha-Glucosidase and Alpha-Amylase Inhibitory Activities and Antioxidant Capacity of Helichrysum cymosum and Helichrysum pandurifolium Schrank Constituents

Diabetes mellitus (DM) is a group of systemic metabolic disorders with high rate morbidity and mortality worldwide. Due to the detrimental side effects current treatment, there great need develop more effective antidiabetic drugs fewer effects. Natural products are well-known source for discovery new scaffolds drug discovery, including drugs. The genus Helichrysum has been shown produce natural products. In this investigation, methanolic extract H. cymosum pandurifolium resulted in isolation identification eleven known compounds viz 5,8-dihydroxy-7-methoxy-2-phenyl flavanone (1), pinostrobin (2), dihydrobaicalein (3), glabranin (4), allopatuletin (5), chalcone (6), helichrysetin (7), 5-hydroxy-3,7-dimethoxyflavone (8), 3,5-dihydroxy-6,7,8-trimethoxyflavone (9), 3-O-methylquercetin (10), 3-methylethergalangin (11). vitro bio-evaluation isolated against alpha-glucosidase showed that 10, 5, 11 demonstrated highest inhibitory activity IC50 values 9.24 ± 0.4, 12.94 0.2, 16.00 2.4 μM respectively, followed by 7 3 18.16 1.2 44.44 0.2 respectively. However, none these measurable effect on alpha-amylase under experimental conditions used except compound 10 which poor an value 230.66 15.8 μM. Additionally, strong total antioxidant capacities were 5 ferric-ion reducing power assay (374.34 69.7; 334.37 1.7; 279.93 0.8) µmol AAE/mmol. This first scientific report be carried out activities constituents methoxyflavanoids from pandurifolium. Our findings suggest promising candidates inhibit as well oxidative stress related diabetes. Results molecular docking provided insight into observed 7, 11. It envisaged phytochemicals plants may contribute development hypoglycemic lead anti-diabetic potential.
