Involvement of 9-O-Acetyl GD3 ganglioside in Mycobacterium leprae infection of Schwann cells.

Mycobacterium leprae (ML), the etiologic agent of leprosy, mainly affects the skin and peripheral nerves, leading to demyelization and loss of axonal conductance. Schwann cells (SCs) are the main cell population infected by ML in the nerves, and infection triggers changes in the SC phenotype from a myelinated to a nonmyelinated state. In the present study, we show that expression of 9-O-acetyl ...

Structure of Mycobacterium leprae.

Introduction : why study structure? Information about the structure of M. leprae may be obtained by microscopy (especially electron miscroscopy) and other physical techniques . There is active development of such techniques , but at present the information they can give about structures as complex as a bac terium is l imited . To obtain a detailed knowledge of the molecular composition and con...

Ubiquilin 1 Promotes IFN-γ-Induced Xenophagy of Mycobacterium tuberculosis

The success of Mycobacterium tuberculosis (Mtb) as a pathogen rests upon its ability to grow intracellularly in macrophages. Interferon-gamma (IFN-γ) is critical in host defense against Mtb and stimulates macrophage clearance of Mtb through an autophagy pathway. Here we show that the host protein ubiquilin 1 (UBQLN1) promotes IFN-γ-mediated autophagic clearance of Mtb. Ubiquilin family members ...

Adjuvant activity of Mycobacterium leprae.

Mycobacterium leprae organisms isolated from infected spleen and liver tissue by zonal centrifugation were shown to possess adjuvant activity. Histochemical examination of the footpad macrophage epithelioid granuloma showed that macrophages contained large amounts of hemosiderin after the injection of M. leprae and much smaller amounts after the injection of M. tuberculosis.

Involvement of glutathione in the cytotoxicity of 9-isothiocyanatoacridine.

Isothiocyanates (ITCs) are phytochemicals with promising cancer-preventive potential. To elucidate the mechanism of cytotoxicity of ITCs, their accumulation by cells and the role of intracellular glutathione, fluorescent 9-isothiocyanatoacridine (AcITC) was synthesized. The kinetic parameters for the reactions of AcITC with thiols were estimated and the influence of AcITC on human chronic myelo...
