Drosophila USP22/nonstop polarizes the actin cytoskeleton during collective border cell migration

چکیده

Polarization of the actin cytoskeleton is vital for collective migration cells in vivo. During invasive border cell Drosophila, polarization directly controlled by Hippo signaling complex, which resides at contacts between cluster. Here, we identify, a genetic screen deubiquitinating enzymes involved migration, an essential role nonstop/USP22 expression pathway components expanded and merlin. Loss nonstop function consequently leads to redistribution F-actin polarity determinant Crumbs, loss polarized protrusions, tumbling Nonstop component Spt-Ada-Gcn5-acetyltransferase (SAGA) transcriptional coactivator but SAGA’s histone acetyltransferase module, does not bind or merlin, dispensable migration. Taken together, our results uncover novel roles SAGA-independent may help guide studies other systems where USP22 necessary motility invasion.

The Hippo pathway polarizes the actin cytoskeleton during collective migration of Drosophila border cells

Collective migration of Drosophila border cells depends on a dynamic actin cytoskeleton that is highly polarized such that it concentrates around the outer rim of the migrating cluster of cells. How the actin cytoskeleton becomes polarized in these cells to enable collective movement remains unknown. Here we show that the Hippo signaling pathway links determinants of cell polarity to polarizati...

Ste20/GCK kinase Nak1/Orb3 polarizes the actin cytoskeleton in fission yeast during the cell cycle.

Polar growth is a crucial process during cell morphogenesis. The microtubule and actin cytoskeletons, and vesicular transport are tightly regulated to direct cellular growth and to generate specific cell forms. We demonstrate here that the Ste20-related protein kinase Nak1/Orb3 is required in fission yeast to polarize the actin cytoskeleton at the tips of the cells and for cell separation, and ...

Mitochondrial Ca2+ uptake controls actin cytoskeleton dynamics during cell migration

Intracellular Ca2+ signaling regulates cell migration by acting on cytoskeleton architecture, cell directionality and focal adhesions dynamics. In migrating cells, cytosolic Ca2+ pool and Ca2+ pulses are described as key components of these effects. Whereas the role of the mitochondrial calcium homeostasis and the Mitochondria Cacium Uniporter (MCU) in cell migration were recently highlighted i...

Tousled-like kinase regulates cytokine-mediated communication between cooperating cell types during collective border cell migration

Collective cell migration is emerging as a major contributor to normal development and disease. Collective movement of border cells in the Drosophila ovary requires cooperation between two distinct cell types: four to six migratory cells surrounding two immotile cells called polar cells. Polar cells secrete a cytokine, Unpaired (Upd), which activates JAK/STAT signaling in neighboring cells, sti...

Cell-cell communication during collective migration.

Cell motility plays a critical role in many biological and medical processes, including wound healing, morphogenesis, and cancer metastasis (1). Often, this movement is guided by external chemical cues in the form of chemoattractant gradients. These cues consist of diffusive chemoattractant molecules that bind to surface receptors on the cell membrane and activate intracellular signaling pathwa...
