Human Indoleamine 2,3-dioxygenase 1 (IDO1) Expressed in Plant Cells Induces Kynurenine Production

Genetic engineering of plants has turned out to be an attractive approach produce various secondary metabolites. Here, we attempted kynurenine, a health-promoting metabolite, in Nicotiana tabacum (tobacco) transformed by Agrobacterium tumefaciens with the gene, coding for human indoleamine 2,3-dioxygenase 1 (IDO1), enzyme responsible kynurenine production because tryptophan degradation. The presence IDO1 gene transgenic was confirmed PCR, but protein failed detected. To confer higher stability heterologous and provide more sensitive method detect interest, cloned construct IDO1-GFP. Analysis transiently transfected tobacco protoplasts demonstrated that IDO1-GFP led expression detectable protoplast medium. Interestingly, intracellular localisation plant cells is similar found mammal cells, mainly cytosol, early endosomes as well. best our knowledge, this first report on capable secreting kynurenines cells.
