HMGB1-activated fibroblasts promote breast cancer cells metastasis via RAGE/aerobic glycolysis

چکیده
Highly expressed high mobility group box-1 protein (HMGB1) promotes tumor metastasis. Whether HMGB1 participates in breast cancer cell activation of fibroblasts is unknown. The culture medium 6 lines with different migration potential, and overexpression or knockdown was used to induce fibroblast activation, collagen α-SMA expression were measured. We evaluated the potential MDA-MB-231 cells treated 3-PO (3-(3-pyridinyl)-1-(4-pyridinyl)-2-propen-1-one) inhibitor, anti-HMGB1 treatment, RAGE (receptor for advanced glycation end products) knockdown. A lung metastasis murine model evaluate whether RAGE-knockdown mitigates Breast that are highly migratory have a invasive had higher induced greater strongly than poorer motility. hrHMGB1 supernatants HMGB1-overexpressed MCF-7 promoted but loss-HMGB1 abolished potential. Moreover, novel mechanism identified by which facilitated RAGE/aerobic glycolysis. Consistently, enhanced metastasis, enhancement reversed inhibition, vitro vivo. secreted via glycolysis, activated enhance through increased lactate.

Tissue-resident stem cells promote breast cancer growth and metastasis.
Mesenchymal stem cells derived from bone marrow have recently been described to localize to breast carcinomas and to integrate into the tumor-associated stroma. In the present study, we investigated whether adipose tissue-derived stem cells (ASCs) could play a role in tumor growth and invasion. Compared with bone marrow-derived cells, ASCs as tissue-resident stem cells are locally adjacent to b...
متن کاملBreast Cancer Metastasis to Pituitary Infandibulum
Metastasis from breast cancer to other parts of the body is very common, but the spread of the tumor to pituitary gland, especially to infandibulum, is a rare presentation. At the time of pituitary metastasis, a majority of the patients have clinical and radiological evidence of the disease. It seems that the posterior area of the gland is the most common site of metastasis, probably due to hig...
متن کاملBreast cancer cells condition lymphatic endothelial cells within pre-metastatic niches to promote metastasis
Breast cancer metastasis involves lymphatic dissemination in addition to hematogenous spreading. Although stromal lymphatic vessels (LVs) serve as initial metastatic routes, roles of organ-residing LVs are underinvestigated. Here we show that lymphatic endothelial cells (LECs), a component of LVs within pre-metastatic niches, are conditioned by triple-negative breast cancer (TNBC) cells to acce...
متن کاملStromal fibroblasts activated by tumor cells promote angiogenesis in mouse gastric cancer.
Myofibroblasts, also known as activated fibroblasts, constitute an important niche for tumor development through the promotion of angiogenesis. However, the mechanism of stromal fibroblast activation in tumor tissues has not been fully understood. A gastric cancer mouse model (Gan mice) was recently constructed by simultaneous activation of prostaglandin (PG) E2 and Wnt signaling in the gastric...
متن کاملCancer-Associated Fibroblasts Promote Proliferation of Endometrial Cancer Cells
Endometrial cancer is the most commonly diagnosed gynecologic malignancy worldwide; yet the tumor microenvironment, especially the fibroblast cells surrounding the cancer cells, is poorly understood. We established four primary cultures of fibroblasts from human endometrial cancer tissues (cancer-associated fibroblasts, CAFs) using antibody-conjugated magnetic bead isolation. These relatively h...
