Abstract Chimeric antigen receptor (CAR)-T cell therapy is an antigen-dependent cellular that has gained considerable traction in the field of cancer immunotherapy. Currently, there are six FDA-approved CAR-T therapies, which all target CD19 or BCMA antigens for hematologic B malignancies. In recent years, a strong focus been placed on CAR T discovery solid tumors, may better recapitulate physiological conditions, thereby potentially improving selection construct candidates. Therefore, it critical to develop novel methodologies high-throughput phenotypic and functional assays using 3D tumor spheroid models assess therapies against tumors. Recently, plate-based image cytometry emerged as method investigate characterize functions manner.
