Heterochromatic Repeat Clustering Imposes a Physical Barrier on Homologous Recombination to Prevent Chromosomal Translocations

چکیده

Mouse pericentromeric DNA is composed of tandem major satellite repeats, which are heterochromatinized and cluster together to form chromocenters containing homologous sequences from different chromosomes. These clusters protected damage refractory repair through recombination (HR). Moreover, double stranded breaks (DSBs) occurring within relocate away the core domain be repaired. The mechanisms by pericentric heterochromatin imposes a barrier on HR implications repeat clustering unknown. Here, we compare spatial recruitment factors upon DSBs induced specifically in human mouse heterochromatin, differ their capacity clusters. We show that while increase accessibility disrupting HP1 α dimerization, physical requires many layers decompaction accessed. Our results consistent with model 3D organization heterochromatic regions dictates activation pathways key prevent clustered repeats onset chromosomal translocations.

Telomerase Deficiency Affects the Formation of Chromosomal Translocations by Homologous Recombination in Saccharomyces cerevisiae

Telomerase is a ribonucleoprotein complex required for the replication and protection of telomeric DNA in eukaryotes. Cells lacking telomerase undergo a progressive loss of telomeric DNA that results in loss of viability and a concomitant increase in genome instability. We have used budding yeast to investigate the relationship between telomerase deficiency and the generation of chromosomal tra...

Balanced Chromosomal Translocations of Parents in Relation to Spontaneous Abortions

The most significant complication of pregnancy is recurrent miscarriage. Numerous factors have been described as associations with recurrent wastage such as: uterine abnormalities, immunological factors, endocrinologic imbalance and chromosomal defects. Cytogenetic evaluation of couples with recurrent pregnancy losses is performed on the basis of G-banding technique only after other possible et...

Switching on chromosomal translocations.

Activation-induced deaminase initiates three different antibody diversification reactions: class switch recombination, somatic hypermutation (SHM), and gene conversion. We have shown that, in addition to antibody diversification, activation-induced deaminase can also initiate Burkitt's lymphoma-like c-myc/IgH translocations. However, distinct DNA damage- and oncogene-induced checkpoints operate...

Designing E1 Deleted Adenoviral Vector by Homologous Recombination

Adenoviruses are used extensively to deliver genes into mammalian cells, particularly where there is a requirement for high-level expression of transgene products in cultured cells, or for use as recombinant viral vaccines or in gene therapy. In spite of their usefulness, the construction of adenoviral vectors (AdV) is a cumbersome and lengthy process that is not readily amenable to the generat...
