چکیده
Abstract Hepatitis B–associated hepatocellular carcinoma (HCC) is often accompanied by severe vascular invasion and portal vein tumor thrombus, leading to a poor prognosis. However, the underlying mechanism of this disease remains obscure. In study, we demonstrate that hepatitis B virus (HBV)–encoded gene HBx induces high IL8 production through MEK–ERK signal activation, enhanced endothelial permeability facilitate invasion. metastatic model using tail injection in transgenic mouse with selective expression human CXCR1 endothelium, activation IL8–CXCR1 cascade overexpression cells dramatically liver metastasis. Mechanistically, selectively induced GARP-latent-TGFβ sinusoidal subsequently provoked preferential regulatory T-cell polarization suppress antitumor immunity. Collectively, these findings reveal signaling axis mediates local microenvironmental immune escape HCC induce intrahepatic metastasis, which may serve as potential therapeutic targets for HBV-associated HCC. Significance: This study identifies B–induced IL8/CXCR1/TGFβ suppresses immunity enhances metastasis carcinoma, providing new intervention.

14-3-3ζ promotes hepatocellular carcinoma venous metastasis by modulating hypoxia-inducible factor-1α
Portal vein tumor thrombus (PVTT) is a type of intrahepatic metastasis arising from hepatocellular carcinoma (HCC) and is highly correlated with a poor prognosis. Hypoxia is common in solider tumors, including HCC, where it alters the behavior of HCC cells. We asked whether and how hypoxia contributes to PVTT formation. We demonstrated that increased intratumoral hypoxia is strongly associated ...
متن کاملFNDC3B promotes cell migration and tumor metastasis in hepatocellular carcinoma
Recurrence and metastasis are common in hepatocellular carcinoma (HCC) and correlate with poor prognosis. We investigated the role of fibronectin type III domain containing 3B (FNDC3B) in HCC metastasis. Overexpression of FNDC3B in HCC cell lines enhanced cell migration and invasion. On the other hand, knockdown of FNDC3B using short-hairpin RNA reduced tumor nodule formation in both intra- and...
متن کاملCD146 promotes metastasis and predicts poor prognosis of hepatocellular carcinoma.
BACKGROUND Hepatocellular carcinoma (HCC) is the third leading cause of cancer-related mortality worldwide. Recurrence and metastasis after curative resection remain critical obstacles in HCC treatment. CD146 predicted poor prognosis of a variety of cancers including melanoma, breast tumors, prostate cancer, and gastric cancer. However, the role of CD146 in HCC has not yet been systematically e...
متن کاملAccumulation of 99mTc-PMT in renal metastasis of hepatocellular carcinoma.
We describe here a case in which 99mTc-Sn-N-pyridoxy-5-methyltryptophan (99mTc-PMT) scintigraphy was useful in diagnosing renal metastasis of hepatocellular carcinoma (HCC). A 62-year-old man who had undergone hepatectomy for HCC presented 6 years after initial diagnosis with left flank pain and was found on CT and MRI to have a tumor in the left kidney. Hepatobiliary scintigraphy using 99Tc-PM...
متن کاملRho kinase phosphorylation promotes ezrin-mediated metastasis in hepatocellular carcinoma.
During progression of hepatocellular carcinoma, multiple genetic and epigenetic alterations act to posttranslationally modulate the function of proteins that promote cancer invasion and metastasis. To define such abnormalities that contribute to liver cancer metastasis, we carried out a proteomic comparison of primary hepatocellular carcinoma and samples of intravascular thrombi from the same p...
