Helminth Imprinting of Hematopoietic Stem Cells Sustains Anti-Inflammatory Trained Innate Immunity That Attenuates Autoimmune Disease

چکیده
Abstract Certain proinflammatory stimuli can metabolically and epigenetically modify monocytes/macrophages or NK cells to be more responsive secondary stimuli, a process known as trained innate immunity. However, the longevity of immunity is unclear. In this study, we report that Fasciola hepatica excretory-secretory products (FHES) imprint an anti-inflammatory phenotype on long-term hematopoietic stem (HSCs) monocyte precursor populations, enhancing their proliferation differentiation into Ly6Clow monocytes. These monocytes expand populate multiple compartments within mice, conferring hyporesponsiveness reduced susceptibility induction experimental autoimmune encephalomyelitis. Mice treated with FHES had enhanced alternatively activated macrophages, Th1 Th17 responses, attenuating effects autoimmunity persisted for 8 mo. Furthermore, transplantation HSCs from FHES-treated mice transferred naive mice. Our findings demonstrate helminth modulate promote development myeloid attenuate T cell–mediated disease.

Hematopoietic Stem and Progenitor Cells as Effectors in Innate Immunity
Recent research has shed light on novel functions of hematopoietic stem and progenitor cells (HSPC). While they are critical for maintenance and replenishment of blood cells in the bone marrow, these cells are not limited to the bone marrow compartment and function beyond their role in hematopoiesis. HSPC can leave bone marrow and circulate in peripheral blood and lymph, a process often manipul...

متن کاملgeneration and characterization of anti-cd34 monoclonal antibodies that react with hematopoietic stem cells
cd34 is a type i membrane protein with a molecular mass of approximately 110 kda. this antigen is associated with human hematopoietic progenitor cells and is a differentiation stage-specific leukocyte antigen. in this study we have generated and characterized monoclonal antibodies (mabs) directed against a cd34 marker. mice were immunized with two keyhole lympet hemocyanin (klh)-conjugated cd34...
متن کاملInflammatory bowel disease related innate immunity and adaptive immunity.
Inflammatory bowel disease (IBD) is a chronic nonspecific intestinal inflammatory disease, including ulcerative colitis (UC) and Crohn's disease (CD). Its pathogenesis remains not yet clear. Current researchers believe that after environmental factors act on individuals with genetic susceptibility, an abnormal intestinal immune response is launched under stimulation of intestinal flora. However...
