Genomic Instability in Somatic Hybridization between Poncirus and Citrus Species Aiming to Create New Rootstocks

چکیده
Rootstocks are an important component for citrus adaptation to increasing biotic and abiotic stresses resulting from global climate change. There is a strong complementarity between Citrus species, which adapt stresses, Poncirus trifoliata its intergeneric hybrids, exhibit resistances or tolerances major diseases pests. Thus, symmetrical somatic hybridization complementary diploid rootstocks of these two genera appears be efficient way develop new tetraploid in order address the challenges industry. New hybrids were obtained by electrofusion protoplasts P. hybrids. Extensive characterization nuclear cytoplasmic genomes was performed genotyping-by-sequencing (GBS) analysis. This revealed cybrids Mitochondrial mostly inherited callus parent, but homologous recombination events observed one parental combination. Chloroplasts exhibited random uniparental inheritance. GBS local chromosomal instabilities all whole chromosome eliminations However, at genome level, addition both parents predominant displayed least trifoliate orange haplotype throughout genome.

Plant growth and K influx responses to K supply were studied in two citrus rootstocks (Carrizo citrange, Citrus sinensis [L.] Osb. × Poncirus trifoliata [L.] Raf.; Cleopatra mandarin, Citrus reshni Hort. ex Tanaka) to determine mechanisms of K acquisition. Seedlings of Carrizo citrange (CC) and Cleopatra mandarin (Cleo) were grown in aerated nutrient solutions for 40 days with either no K added...

The response of Citrus spp. and related rootstocks to a population of Meloidogyne javanica was evaluated in a screenhouse experiment. Palestine and Rangpur lime, rough lemon, sour orange, Sexton and Thentriton tangelo, and Volkamer lemon were not infected by M. javanica. Galls and tip swellings were observed on the roots of Poncirus triloliata and Troyer citrange. There was no evidence of nemat...

In situ hybridization of 18S-5.8S-25S rDNA probes labeled with biotin or rhodamine and 5S rDNA probes labeled with digoxigenin was used to locate rDNA sites on root-tip metaphase chromosomes of Citrus sinensis L. (2n = 2x = 18), Poncirus trifoliata L. Raf. (2n = 2x = 18), and Citrus x Poncirus hybrids (2n = 2x = 18). Counterstaining with the fluorochromes chromomycin A3 and DAPI uniquely identi...

Two citrus rootstocks, Rangpur lime (RL) and Sunki Maravilha mandarin (SM), were analyzed either ungrafted or grafted with their reciprocal graft combinations or with shoot scions of two commercial citrus varieties: Valencia orange (VO) and Tahiti acid lime (TAL). All graft combinations were subjected to distinct watering regimes: well-watered, severe drought and rehydration. Growth and water r...

Hybrids as various resources have valuable importance in tree breeding. Today, improvement programs by using poplar and willows and their hybrids play basic role in supplying wood and decreasing pressure on natural forests. This research was carried out in order to study inter-generic hybridization possibility between Salix aegyptica and Populus caspica species from Salicaceae to achieve new hy...
