Genomic Characterization of Multidrug-Resistant Carbapenemase-Producing Enterobacter cloacae ECL189, Co-producing KPC-2, NDM-1, TEM-1, TEM-95, and SHV-66

Background: Carbapenem-resistant Enterobacteriaceae (CRE) has become a public health threat due to resistance multiple antibiotics. The production of β-lactamase is the most important mechanism Enterobacteriaceae. Although isolates producing KPC-2 or NDM-1 enzymes have been reported widely, co-producing KPC-2, NDM-1, TEM-1, TEM-95, SHV-66, and other β-lactamases rarely detected in same strain, especially Enterobacter cloacae. Objectives: In this study, we identified sequenced genome carbapenem-resistant E. cloacae ECL189 in-depth analyze transmission mechanisms Methods: We investigated antimicrobial susceptibility by VITEK 2 system, E-test gradient strips, K-B method. Whole-genome sequencing was used PacBio RS II platform Illumina HiSeq 4000 platform. Antimicrobial genes, virulence non-coding RNA, repeat sequences were predicted biological information databases. A PCR further confirm that blaKPC-2, blaNDM-1, blaTEM-1, blaTEM-95, blaSHV-66 genes existed ECL189. conjugation experiment performed determine transferability resistance. Molecular typing done multilocus sequence (MLST). Results: resistant 21 out 23 tested antibiotics, but its transconjugant 10 18 consisted 5,026,406 bp chromosome four circular plasmids. total, 26 58 proteins identified. addition, 77 determinants associated with bacterial large number located results whole-genome consistent genes. MLST analysis revealed strain belonged ST74. Conclusions: This study resistance, virulence, Resistance spread bacteria horizontal transfer plasmids, which should attract more attention relevant departments.
