Genome-wide clonal variability in European pear “Rocha” using high-throughput sequencing

چکیده
Abstract Pears (Pyrus) are one of the most economically important fruits worldwide. The Pyrus genus is characterized by a high degree genetic variability between species and interspecific hybrids, several studies have been performed to assess this for both cultivated wild accessions. These mostly limited resolving power traditional molecular markers, although in recent past availability reference genome sequences or SNP arrays pear enhanced capability high-resolution genomics studies. tools can also be applied better understand intra-varietal (or clonal) pear. Here we report first resolution analysis clonal population using whole sequencing (WGS). Results showed unique signatures accumulation mutations transposable element insertions each clone, which likely related their history propagation cultivation. nucleotide diversity remained low collection with exception few genomic windows, suggesting that balancing selection may occurring. windows included mainly genes plant fertility. Regions higher mutational load were partially associated transcription factors, probably reflecting distinctive phenotypes collection. annotation variants revealed theoretical disruption relevant Taken together, results from study show clones accumulate differently, those play role on phenotypes, meaning populations studies, when comparing association

Pilot genome-wide study of tandem 3′ UTRs in esophageal cancer using high-throughput sequencing
Regulatory regions within the 3' untranslated region (UTR) influence polyadenylation (polyA), translation efficiency, localization and stability of mRNA. Alternative polyA (APA) has been considered to have a key role in gene regulation since 2008. Esophageal carcinoma is the eighth most common type of cancer worldwide. The association between polyA and disease highlights the requirement for com...
متن کاملHigh-throughput sequencing of the melanoma genome.
Next-generation sequencing technologies are now common for whole-genome, whole-exome and whole-transcriptome sequencing (RNA-seq) of tumors to identify point mutations, structural or copy number alterations and changes in gene expression. A substantial number of studies have already been performed for melanoma. One study analysed eight melanoma cell lines with RNA-Seq technology and identified ...
متن کاملSNP Discovery in European Anchovy (Engraulis encrasicolus, L) by High-Throughput Transcriptome and Genome Sequencing
Increased throughput in sequencing technologies has facilitated the acquisition of detailed genomic information in non-model species. The focus of this research was to discover and validate SNPs derived from the European anchovy (Engraulis encrasicolus) transcriptome, a species with no available reference genome, using next generation sequencing technologies. A cDNA library was constructed from...
متن کاملGenome-Wide Identification of Bone Metastasis-Related MicroRNAs in Lung Adenocarcinoma by High-Throughput Sequencing
BACKGROUND MicroRNAs (miRNAs) are a class of small noncoding RNAs that regulate gene expression at the post-transcriptional level. They participate in a wide variety of biological processes, including apoptosis, proliferation and metastasis. The aberrant expression of miRNAs has been found to play an important role in many cancers. RESULTS To understand the roles of miRNAs in the bone metasta...
