Live attenuated bacteria have been used as target vehicles for genetic therapy of malignant carcinoma because they can be reprogrammed by following simple rules and the ability to tumor hypoxic region. In this research, noninvasive Escherichia coli (E. Coli) is genetically modified through plasmid transfection afford E. Coli(p) with overexpressed human catalase catalyzing H2O2 into O2 in site. The produced consequently converted cytotoxic 1O2 under near-infrared (NIR) light irradiation photodynamic therapy. Chlorin e6 (Ce6) chosen photosensitizer its excellent ability, polydopamine (pDA) employed encapsulate Ce6 good biosafety, photothermal adhesion capacity bacteria. Dopamine polymerizes presence form pDA/Ce6, then coated pDA/Ce6 final Coli(p)/pDA/Ce6. obtained system intravenously administrated selective accumulation replication tumor. NIR introduced enable O2-enhanced On account complementary combination, exhibits efficient antitumor effect vitro vivo.
