چکیده
Complement C1q is a multifunctional protein able to sense pathogens and immune molecules such as immunoglobulins pentraxins, trigger the classical complement pathway through activation of its two associated proteases, C1r C1s. multimeric composed three homologous yet distinct polypeptide chains A, B, C, each an N-terminal collagen-like sequence C-terminal globular gC1q module, that assemble into six heterotrimeric (A-B-C) subunits. This hexameric structure exhibits characteristic shape bouquet flowers, comprising triple helices, terminating in trimeric head. We have produced previously functional recombinant full-length stably transfected HEK 293-F cells, with FLAG tag inserted at end C1qC chain. report here generation additional proteins, fused C-terminus C1qA or C1qB chains, N-terminus Two other variants harboring Myc 6-His were also produced. show all variants, except for His-tagged protein, can be comparable yields are bind similar affinities either IgM, ligand regions, C1r2-C1s2 tetramer, IgM-mediated serum activation. These new provide tools investigate multiple functions C1q.

Expression of recombinant human complement C1q allows identification of the C1r/C1s-binding sites.
Complement C1q is a hexameric molecule assembled from 18 polypeptide chains of three different types encoded by three genes. This versatile recognition protein senses a wide variety of immune and nonimmune ligands, including pathogens and altered self components, and triggers the classical complement pathway through activation of its associated proteases C1r and C1s. We report a method for expr...
متن کاملThree Recombinant Engineered Antibodies against Recombinant Tags with High Affinity and Specificity
We describe three recombinant engineered antibodies against three recombinant epitope tags, constructed with divalent binding arms to recognize divalent epitopes and so achieve high affinity and specificity. In two versions, an epitope is inserted in tandem into a protein of interest, and a homodimeric antibody is constructed by fusing a high-affinity epitope-binding domain to a human or mouse ...
متن کاملIncreased Complement C1q Level Marks Active Disease in Human Tuberculosis
BACKGROUND Complement functions as an important host defense system and complement C5 and C7 have been implicated in immunopathology of tuberculosis. However, little is known about the role of other complement components in tuberculosis. METHODS Complement gene expression in peripheral blood mononuclear cells of tuberculosis patients and controls were determined using whole genome transcripti...
متن کاملComplement, C1q, and C1q-Related Molecules Regulate Macrophage Polarization
Complement is a critical system of enzymes, regulatory proteins, and receptors that regulates both innate and adaptive immune responses. Natural mutations in complement molecules highlight their requirement in regulation of a variety of human conditions including infectious disease and autoimmunity. As sentinels of the immune system, macrophages are specialized to respond to infectious microbes...
متن کاملTrichinella spiralis Paramyosin Binds Human Complement C1q and Inhibits Classical Complement Activation
BACKGROUND Trichinella spiralis expresses paramyosin (Ts-Pmy) as a defense mechanism. Ts-Pmy is a functional protein with binding activity to human complement C8 and C9 and thus plays a role in evading the attack of the host's immune system. In the present study, the binding activity of Ts-Pmy to human complement C1q and its ability to inhibit classical complement activation were investigated. ...
