Fluorescence imaging of chromosomal DNA using click chemistry

Chromosome visualization is essential for chromosome analysis and genetic diagnostics. Here, we developed a click chemistry approach for multicolor imaging of chromosomal DNA instead of the traditional dye method. We first demonstrated that the commercially available reagents allow for the multicolor staining of chromosomes. We then prepared two pro-fluorophore moieties that served as light-up ...

Site-specific incorporation of diamondoids on DNA using click chemistry.

A simple and robust solid phase synthetic method for the ligation of diamondoids on the phosphate backbone of DNA with "click" chemistry using [Cu(CH(3)CN)(4)]PF(6) without a stabilizing ligand is reported. It was found that as the size of dimondoid increased, a corresponding increase in melting temperature was observed.

Imaging Cell Surface Glycosylation in Vivo Using “Double Click” Chemistry

Dynamic alterations in cell surface glycosylation occur in numerous biological processes that involve cell-cell communication and cell migration. We report here imaging of cell surface glycosylation in live mice using double click chemistry. Cell surface glycans were metabolically labeled using peracetylated azido-labeled N-acetylgalactosamine and then reacted, in the first click reaction, with...

Quantification of DNA through a fluorescence biosensor based on click chemistry.

A simple, sensitive and selective fluorescence biosensor for determination of DNA using CuS particles based on click chemistry is reported. Biotin-modified capture DNA was modified on Streptavidin MagneSphere Paramagnetic Particles (PMPs) and hybridized with target DNA (hepatitis B virus DNA had been chosen as an example), then bound target DNA was hybridized with DNA-CuS particles and formed a...

Covalent, sequence-specific attachment of long DNA molecules to a surface using DNA-templated click chemistry.

We present a novel method that covalently and sequence-specifically attaches long DNA molecules to a surface that is compatible with high-resolution atomic force microscopy (AFM) imaging. Surfaces prepared with this approach are ideally suited for performing biophysical experiments on single DNA molecules.
