چکیده
The genus Begomovirus (family Geminiviridae) contains pathogens that infect economically important crops, including cassava, cotton, pepper, potato and tomato (Tabein et al., 2021). During 2019–2021, two hibiscus plants with begomovirus-like symptoms vein enation cup-shaped leaves (Figs. 1, 2) were observed in the botanical garden of Shahid Chamran University Ahvaz, Khuzestan Province, southwestern Iran. New obtained from cuttings diseased showed same (Fig. 3). To investigate cause disease, evaluated using RNA-Seq approach. Total RNA was extracted Column isolation kit (Denazist, Iran) according to manufacturer's instructions. library ready after end repair, adapter ligation, size selection, amplification purification TruSeq Stranded Library Preparation Kit Ribo-Zero depletion. Sequencing performed Illumina´s NovaSeq 6000 (Novogene, China) 150 bp length 6G data depth. Reads (41,872,710 reads) trimmed for adapters, ambiguous nucleotides low quality CLC genomics workbench 20 (Qiagen, Germany), default parameters: bases below 15 nt, maximum 2 Qscore ≤5 trimmed. Clean reads (41,849,488 mapped reference genome (GCA_019671005), then unmapped (3,321,246 assembled. In South Asia, Cotton leaf curl disease (CLCuD) is caused by a complex of phylogenetically-related begomovirus species and a specific betasatellite, Cotton leaf curl Multan betasatellite (CLCuMuB). The post-transcriptional gene silencing (PTGS) suppression activities of the transcriptional activator protein (TrAP), C4, V2 and βC1 proteins encoded by Cotton leaf curl Kokhran virus (CLCuKoV)/CLCu...
متن کاملAn Insight into Cotton Leaf Curl Multan Betasatellite, the Most Important Component of Cotton Leaf Curl Disease Complex
Cotton leaf curl disease (CLCuD) is one of the most economically important diseases and is a constraint to cotton production in major producers, Pakistan and India. CLCuD is caused by monopartite plant viruses belonging to the family Geminiviridae (genus Begomovirus), in association with an essential, disease-specific satellite, Cotton leaf curl Multan betasatellite (CLCuMuB) belonging to a new...
متن کاملMaintenance of Cotton Leaf Curl Multan Betasatellite by Tomato Leaf Curl New Delhi Virus—Analysis by Mutation
Viruses of the genus Begomovirus (family Geminiviridae) are economically important phytopathogens that are transmitted plant-to-plant by the whitefly Bemisia tabaci. Most Old World (OW) begomoviruses are monopartite and many of these interact with symptoms and host range determining betasatellites. Tomato leaf curl New Delhi virus (ToLCNDV) is one of only a few OW begomoviruses with a bipartite...
متن کاملEngineered Disease Resistance in Cotton Using RNA-Interference to Knock down Cotton leaf curl Kokhran virus-Burewala and Cotton leaf curl Multan betasatellite Expression
Cotton leaf curl virus disease (CLCuD) is caused by a suite of whitefly-transmitted begomovirus species and strains, resulting in extensive losses annually in India and Pakistan. RNA-interference (RNAi) is a proven technology used for knockdown of gene expression in higher organisms and viruses. In this study, a small interfering RNA (siRNA) construct was designed to target the AC1 gene of Cott...

Cotton leaf curl disease (CLCuD) in Pakistan and northwestern India is caused by monopartite begomoviruses in association with an essential, disease-specific satellite, Cotton leaf curl Multan betasatellite (CLCuMB). Following a recent upsurge in CLCuD problems in Sindh province (southern Pakistan), sequences of clones of CLCuMB were obtained from Sindh and Punjab province (central Pakistan), w...
