چکیده

The main efforts in common wheat (Triticum aestivum L.) breeding focus on yield, grain quality, and resistance to biotic abiotic stresses. One of the major threats affecting global cultivation causing significant crop production losses are rust diseases, including leaf caused by a biotrophic fungus Puccinia triticina Eriks. Genetically determined has been characterized young plants (seedling resistance) as well at adult plant stage. At seedling stage, is controlled vertically R genes, conferring race-specific response that highly effective but usually short-lived due rapid evolution potentially virulent fungi. In mature plants, horizontal (APR) was described, which provides long-term protection against multiple races pathogens. A better understanding molecular mechanisms underlying function APR genes would enable development new strategies for wheat. Therefore, present study we focused early transcriptomic responses two Lr34 Lr67, three complementary miRNAs, tae-miR9653b, tae-miR9773 tae-miR9677b, inoculation with P. triticina. Plant material consisted five reference varieties, Artigas, NP846, Glenlea, Lerma Rojo TX89D6435, containing Lr34/Yr18 Lr67/Yr46 genes. Biotic stress induced fungal spores under conditions phytotron. tissue sampled before 6, 12, 24 48 h postinoculation (hpi). gene expression quantified using real-time PCR whereas miRNA droplet digital PCR. This paper describes rust-causing fungi occur central Europe. revealed high variability profiles between varieties time-points, prevalence downregulation upregulation miRNAs during an defense response. Nevertheless, despite initially observed, Lr67 studied cultivars significantly higher than control line carrying wild (susceptible) alleles.

Isolation and characterization of ERECTA genes and their expression patterns in common wheat (Triticum aestivum L.)

The orthologue of Arabidopsis ERECTA gene (ER) in wheat, TaER, is considered to be a promising candidate gene for the genetic improvement of water use efficiency (WUE) and drought tolerance in breeding programs. In this study, we isolated two distinct homologues (TaER1 and TaER2) of TaER genes in common wheat through in silico screening and PCR-based homologous cloning. Sequence analysis reveal...

Quantitative expression analysis of candidate genes for Septoria tritici blotch resistance in wheat (Triticum aestivum L.)

Septoria tritici blotch (STB), caused by the ascomycete fungus Mycosphaerella graminicola (asexual stage: Septoria tritici), is one of the most important foliar diseases of wheat. In this research, quantitative expression analysis of five candidate genes for the induction of resistance to STB (PR-1, Bsi, Msr, Per, and Ppi) was conducted in the wheat cultivars ‘Seri 82’ (susceptible) and ‘Fronta...

Ca2+/Cation Antiporters (CaCA): Identification, Characterization and Expression Profiling in Bread Wheat (Triticum aestivum L.)

The Ca2+/cation antiporters (CaCA) superfamily proteins play vital function in Ca2+ ion homeostasis, which is an important event during development and defense response. Molecular characterization of these proteins has been performed in certain plants, but they are still not characterized in Triticum aestivum (bread wheat). Herein, we identified 34 TaCaCA superfamily proteins, which were classi...

Characterization and Expression of High Temperature Stress Responsive Genes in Bread Wheat (Triticum aestivum L.)

To elucidate the effects of high temperatures, wheat plants (Triticum aestivum cv. CPAN 1676) were given heat shocks at 37°C and 42°C for two hours, and responsive genes were identified through PCR-Select Subtraction technology. Four subtractive cDNA libraries, including three forward and one reverse subtraction, were constructed from three different developmental stages. A total of 5500 ESTs w...

The wheat (Triticum aestivum L.) leaf proteome.

The wheat leaf proteome was mapped and partially characterized to function as a comparative template for future wheat research. In total, 404 proteins were visualized, and 277 of these were selected for analysis based on reproducibility and relative quantity. Using a combination of protein and expressed sequence tag database searching, 142 proteins were putatively identified with an identificat...
