چکیده
Statins have anticancer effects and may be used as agents via drug repositioning. In reverse transcription-quantitative polymerase chain reaction (RT-qPCR) assays, the internal reference gene must not affected by any experimental conditions. As statins exert a wide range of on cells inhibiting mevalonate pathway, it is possible that statin treatment might alter expression housekeeping genes genes, thereby misleading assessment obtained data. Here, we evaluated stability in atorvastatin-treated cancer cell lines. We treated both statin-sensitive statin-resistant lines with atorvastatin at seven different concentrations performed RT-qPCR 15 whose was then assessed using five algorithms. lines, certain dose-dependent line-dependent manner; therefore, caution should exercised when comparing target between cells. Our findings emphasize importance validation analyses treatment-based research.

Regulation of VEGF by mevalonate pathway inhibition in breast cancer
Aminobisphosphonates are used for the treatment of benign and malignant bone disorders. As inhibitors of the mevalonate pathway they exert direct anti-tumor effects in vitro and in preclinical models of bone metastases. Bisphosphonates are thought to have an anti-angiogenic activity as decreased levels of VEGF have been reported in some, although not all patients, following treatment with bisph...
متن کاملInhibition of the mevalonate pathway to override chemoresistance and promote the immunogenic demise of cancer cells
The mevalonate pathway is an attractive target for cancer therapy not only to override multidrug resistance but also to promote the immunogenic demise of malignant cells. Recent data indicate that aminobisphosphonates are superior to statins for the pharmacological manipulation of the mevalonate pathway, since they exert therapeutically relevant effects on both cancer cells and the immune system.
متن کاملNDRG2 Regulates the Expression of Genes Involved in Epithelial Mesenchymal Transition of Prostate Cancer Cells
Background: Metastasis is the main cause of prostate cancer (PCa) death. The inhibitory effect of N-myc downstream-regulated gene 2 (NDRG2) on the invasiveness properties of PCa cells has been demonstrated previously. However, its underlying mechanisms have not yet been investigated. The present study aimed to investigate the effects of NDRG2 overexpression on the expression of genes involved i...
متن کاملIdentification and Validation of Housekeeping Genes for Gene Expression Analysis of Cancer Stem Cells
The characterization of cancer stem cell (CSC) subpopulation, through the comparison of the gene expression signature in respect to the native cancer cells, is particularly important for the identification of novel and more effective anticancer strategies. However, CSC have peculiar characteristics in terms of adhesion, growth, and metabolism that possibly implies a different modulation of the ...
متن کاملThe non-mevalonate pathway of isoprenoids: genes, enzymes and intermediates.
Although the mevalonate pathway had been considered for a long time as the unique source of biosynthetic isoprenoids, an alternative pathway has recently been discovered. The first intermediate, 1-deoxy-D-xylulose 5-phosphate, is assembled by condensation of glyceraldehyde 3-phosphate and pyruvate. A skeletal rearrangement coupled with a reduction step affords the branched-chain polyol, 2C-meth...
