Expression of a synthetic gene encoding the enhanced green fluorescent protein in various Escherichia coli strains

Enhanced Green Fluorescent Protein (eGFP) shows much stronger fluorescence than its ancestor, (GFP), thus has been widely applied as a reporter for biomedical research. In this study, we reported the expression of synthetic codon optimized gene encoding eGFP in Escherichia coli (E. coli). The was cloned into two vectors, pQE30 and pColdII resulting recombinant vectors were transformed E. M15 BL21 De3 RIL plus strains, respectively. levels functional showed temperature dependent pattern, which lowering induction increased amount eGFP. Surprisingly, phenomenon called auto-induction when TOP10 cells carrying grown on Luria Broth plates. robust stability even at room temperature, greatly facilitated purification handling. Mouse polyclonal antibodies conveniently generated against protein. Besides potential application coli, systems here are extremely useful models teaching DNA technology undergraduate level.
