Expression and prognostic significance of CBX2 in colorectal cancer: database mining for CBX family members in malignancies and vitro analyses

Abstract Background The Chromobox (CBX) domain protein family, a core component of polycomb repressive complexes 1, is involved in transcriptional repression, cell differentiation, and program development by binding to methylated histone tails. Each CBX family member plays distinct role various biological processes through their own specific chromatin domains, due differences conserved sequences the proteins. It has been demonstrated that colorectal cancer (CRC) multiple-step evolutionary process, whereas roles CRC remain largely unclear. Methods In present study, expression prognostic significance were systematically analyzed series online databases, including Cancer Cell Line Encyclopedia (CCLE), Oncomine, Human Protein Atlas (HPA), Gene Expression Profiling Interactive Analysis (GEPIA). For vitro verification, we performed cloning, flow cytometry transwell experiments verify proliferation invasion ability cells after knocking down CBX2. Results Most proteins found be highly expressed CRC, but only elevated CBX2 could associated with poor prognosis patients CRC. Further examination was several experiments. overexpressed lines via CCLE database results verified RT-qPCR. Moreover, knockdown significantly suppressed invasion. Furthermore, downregulation promote apoptosis. Conclusions Based on these findings, may function as an oncogene potential biomarker. Thus, association between abnormal initiation deserves further exploration.
