Evaluation of two Fasciola hepatica faecal egg counting protocols in sheep and cattle

چکیده

Fascioliasis causes significant economic losses and is a constant challenge to livestock farmers globally. Fluke faecal egg counts (flukeFECs) are simple, non-invasive method used detect the presence of patent liver fluke infection. Many flukeFEC techniques exist but they vary in complexity, precision accuracy. The objective this study was evaluate recovery capabilities two simple methods at different concentrations ruminant species, using artificially spiked samples. We added Fasciola hepatica eggs sheep cattle faeces 2, 5 10 20 epg utilised Flukefinder® (FF) sedimentation (referred as Becker method) investigate effects methods, species density on recovery. calculated proportion recovered each technique, determined lowest reliable detection threshold method. performance were also compared samples collected from naturally infected animals. egg-spiking revealed that both FF significantly more likely recover than (P < 0.001). Overall, 0.001), importantly has low cattle. kappa coefficient indicated substantial agreement between (0.62, P 0.05) moderate (0.41, 0.05). This demonstrated therefore promising potential for future on-farm diagnostics.

Molecular differentiation of sheep and cattle isolates of Fasciola hepatica using RAPD-PCR

Understanding genetic structure and status of genetic variation of Fasciola hepatica isolates from different hosts, has important implications on epidemiology and effective control of fasciolosis. Random amplified polymorphic DNA (RAPD-PCR) was used to study the genetic variation of F. hepatica in sheep and cattle. DNA was extracted from adult helminthes removed from livers of each infected ani...

Sheep and Fasciola hepatica in Europe: the GLOWORM experience.

Fasciola hepatica infection challenges health, welfare and productivity of small ruminants throughout the world. The distribution of F. hepatica in sheep in Europe is usually scattered and studies are generally concerned with a single area making it difficult to compare results from different environments, climates and management regimes. In order to elucidate the current scenario in terms of p...

Local immune response to experimental Fasciola hepatica infection in sheep.

Macrophages, eosinophils, neutrophils and lymphocyte subpopulations (OvCD5+, OvCD4+, OvCD8+, OvWC1+ and lg+) were identified in sections of hepatic tissue and hepatic lymph nodes from sheep experimentally infected with F. hepatica and necropsied 14, 28, 42 or 56 days post infection. The migratory tunnels produced by juvenile flukes appeared as local areas of necrosis surrounded by infiltrating ...

Prevalence of Liver Fluke (Fasciola hepatica) in Wild Red Deer (Cervus elaphus): Coproantigen ELISA Is a Practicable Alternative to Faecal Egg Counting for Surveillance in Remote Populations

Red deer (Cervus elaphus) are hosts of liver fluke (Fasciola hepatica); yet, prevalence is rarely quantified in wild populations. Testing fresh samples from remote regions by faecal examination (FE) can be logistically challenging; hence, we appraise frozen storage and the use of a coproantigen ELISA (cELISA) for F. hepatica surveillance. We also present cELISA surveillance data for red deer fr...
