چکیده

Abstract Background Acute myeloid leukemia (AML) is a heterogeneous disorder encompassing set of hematopoietic tumors that develop when the precursor cells undergo disproportionate clonal proliferation. Homeobox A 9 ( HOXA9 ) pioneer transcription factor in AML pathogenesis along with its cofactor ecotropic integration site 1 MEIS1 ). Our work aimed to evaluate different expression levels and genes their diagnostic prognostic significance adult Egyptian patients de novo AML. The study was carried out on 91 41 healthy individuals. Bone marrow samples were obtained from both controls then tested by reverse transcription-quantitative polymerase chain reaction assess mRNA studied genes. Results gene significantly elevated compared p < 0.001). There statistically significant positive correlation between patients. However, there no association disease-free survival (DFS) overall (OS) = 0.264 0.351, respectively). Conclusion are highly expressed patients, suggesting interesting pathogenic role They could be used as markers for diagnosis AML, but not disease prognosis.

Molecular and Cellular Pathobiology IDH2 and NPM1 Mutations Cooperate to Activate Hoxa9/Meis1 and Hypoxia Pathways in Acute Myeloid Leukemia

IDH1 and IDH2 mutations occur frequently in acute myeloid leukemia (AML) and other cancers. The mutant isocitrate dehydrogenase (IDH) enzymes convert a-ketoglutarate (a-KG) to the oncometabolite 2-hydroxyglutarate (2-HG), which dysregulates a set of a-KG–dependent dioxygenases. To determine whether mutant IDH enzymes are valid targets for cancer therapy, we created a mouse model of AML in which...

Evaluation of procalcitonin as a biomarker of diagnosis, severity and postoperative complications in adult patients with acute appendicitis

Background :Delay in diagnosis and treatment of acute appendicitis (AA) results in an increased rate of perforation, postoperative morbidity, mortality and hospital length of stay. Several biochemical parameters including white blood cell (WBC) count, C-reactive protein (CRP), interleukin-6 (IL6) and Procalcitonin (PCT) have been used to further improve the clinical diagnosis of AA. The aim o...

Unilateral Ptosis as Initial Manifestation of Acute Myeloid Leukemia

Abstract Background In Acute Myeloid Leukemia (AML), malignant clones of immature myeloid cells (primarily blasts) proliferate, replace bone marrow, circulate in blood and invade other tissues. The unique presentation of unilateral ptosis and unilateral eyelid swelling in AML has been reported during the present study. Case Report A 7-year-old girl was encountered with progressively increa...

Hoxa9 and Meis1 Cooperatively Induce Addiction to Syk Signaling by Suppressing miR-146a in Acute Myeloid Leukemia

The transcription factor Meis1 drives myeloid leukemogenesis in the context of Hox gene overexpression but is currently considered undruggable. We therefore investigated whether myeloid progenitor cells transformed by Hoxa9 and Meis1 become addicted to targetable signaling pathways. A comprehensive (phospho)proteomic analysis revealed that Meis1 increased Syk protein expression and activity. Sy...

Mutated NPM1 in combination with overexpression of Meis1 or Hoxa9 is not sufficient to induce acute myeloid leukemia

BACKGROUND Acute myeloid leukemia (AML) carrying nucleophosmin 1 (NPM1) mutations (NPMc(+)) is regarded as a separate entity of myeloid neoplasms due to its distinct biological and clinical features. However, NPMc(+) alone displays low leukemogenic activity and cooperating events appear crucial for AML to develop. Dysregulation of homeobox genes, such as HOXA9 and MEIS1, is a common transcripti...
