Evaluation of Streptocin SH3, a Bacteriocin produced by Streptococcus sanguinis isolated from Human Dental Plaque

Background: Bacteriocin is a peptidic toxin has many advantages to bacteria in their ecological niche and strong antibacterial activity. Objective: The aim of this study was evaluation bacteriocin using Streptococcus sanguinis isolated from human dental caries. Subjects Methods: Thirty five streptococcus isolates were diagnosed tested for production bacteriocin, then the optimal conditions determined. After that, purification made partially by ammonium sulfate at 95% saturation levels, followed gel filtration chromatography Sephadex G-50 column. Finally, physical characteristics also studied determined stability. Results: Among all streptococcal isolates, S. SH3 isolate with batter antimicrobial activity selected, used further experiments. optimum Todd Hewitt broth (THB) supplemented 1% glucose 37°C 24h under anaerobic inoculum size (1.2 ˟ 109CFU/ml) pH 7, where inhibition zone diameter reached 29 mm against Enterococcus faecalis. purified had high inhibitory pneumoniae, pyogenes, E. faecalis, faecium Leuconostoc mesenteroides. Streptocin keeps its effectiveness within range 3-10, temperature until 80ºC 20 min. Conclusion: produced pathogens stability over wide ranges pH, which makes it good alternative medical food preservation fields
